Knockdown of LAMA3 in Cholangiocarcinoma Cells

Small interfering RNAs (siRNAs) were transfected into CCA cell lines using Lipofectamin RNAiMAX reagent (Thermo Fisher Scientific), according to the manufacturer’s instruction. Cells were seeded in a six-well plate at the density of 1.5 × 105 cells/ well (HuCCA-1) and 2 × 105 cells/ well (KKU-213), or in a 96-well plate at the density of 3,000 cells/ well (HuCCA-1) and 4,000 cells/ well (KKU-213) for 24 h prior to transfection with 15 nM siRNA targeting LAMA3 (siLAMA3) (sc-35789; Santa Cruz Biotechnology). AllStars Negative Control siRNA (1027280; Qiagen) was used as control (siNeg). Transfecting medium was replaced with fresh complete media at 24-h post-incubation. Forty-eight hour-post transfection, efficiency of siLAMA3 was evaluated using qRT-PCR and protein expression profile and cell biological phenotypes were assayed as described below.

Integrin β4 and Laminin Receptor Regulation of Cell Migration and Adhesion

Thirty-six hours prior transfection with 40 nM siRNA targeting integrin β4 (siITGB4) (sc-35678; Santa Cruz Biotechnology, Santa Cruz, CA, USA) using Lipofectamin RNAiMAX reagent (Invitrogen, Carlsbad, CA, USA), HuCCA-1 (2 × 105) cells were seeded in 6-well plate, while KKU-213 (2 × 105) cells were used for transfection with either 20 nM siITGB4 or 25 nM siRNA targeting 37/67 kDa laminin receptor (siLAMR) (sc-35789; Santa Cruz Biotechnology). AllStars Negative Control siRNA (siNeg) (1027280; Qiagen, Valencia, CA, USA) was employed as control. Following 24 hours of transfection, cells were incubated with fresh complete media. Migratory and adhesive abilities of cells induced by laminin were determined at 48-hour post-transfection as described below.
