Nitric Oxide Production Measurement

Supernatants from cells treated with BiV for 24 hours were collected to determine NO production, through measurement of nitrite/nitrate levels, using a commercial kit (Roche, Mannheim, Germany). Samples were treated with nitrate reductase (5.7 U/mL) for 30 minutes and then submitted to Griess reaction [28 ]. Additionally, the cells were treated with 25 μM L-NAME (Nω-Nitro-L-arginine methyl ester, Sigma-Aldrich) for 2 hours before the treatment with BiV [29 ], to evaluate the efficacy of NO production blockage by L-NAME.

Essential Oil Analgesic Effects in Mice

All solvents were chromatographic grade (Tedia, Rio de Janeiro, RJ, Brazil). Carrageenan, glutamic acid, atropine, Nω-nitro-L-arginine methyl ester (L-NAME) were purchased from Sigma-Aldrich (St. Louis, MO, USA). Formalin was purchased from Merck (Darmstadt, Germany). Cristália (São Paulo, Brazil) kindly provided morphine sulphate and naloxone hydrochloride. Capsaicin was purchased from Galena (Campinas, SP, Brazil). A stock solution at 100 mg/mL in extrapure oil was prepared with the essential oil (EO). This EO was administered to mice by oral gavage, at doses of 10 to 100 mg/kg, in a final volume of 0.1 mL, 60 min prior to experiments. Morphine (5 mg/kg, p.o.) was diluted in extrapure oil just before use and was used as a reference drug. The control group received vehicle (extrapure oil) by oral gavage.

Historesin is obtained from Leica Biosystems, Wetzlar, Germany, and acetic acid and glucose were obtained from Vetec, Duque de Caxias, Rio de Janeiro, Brazil. Ethyl ether, formaldehyde, and phenolphthalein are from FMaia, Cotia, São Paulo, Brazil; xylazine and ketamine are obtained from Vetbrands, Paulínia, São Paulo, Brazil. Ki-67 protein (code: sc-23900) and BrdU protein (code: sc-32323) are from Santa Cruz Biotechnology, Santa Cruz, CA, USA.

Endothelial Function Evaluation in Rat Aorta

Evaluating Extract's Effect on Uterus

Assessing Oxidative Stress in Diabetic Embryonic Hearts

Hearts collected from E12.5 fetuses from all 4 groups were cryosectioned (CM1950; Leica, Germany) into 8‐μm‐thick slices and placed onto slides. A subset of cryosectioned embryonic hearts from untreated and sapropterin‐treated diabetic dams was incubated with either 300 μmol/L Nω‐nitro‐l‐arginine methyl ester (Sigma), a NOS inhibitor, or 100 U/mL superoxide dismutase (Sigma), a recombinant antioxidant enzyme for 30 minutes. Samples were then probed with 2 μmol/L dihydroethidium (Invitrogen Life Technologies, Burlington, ON, Canada) for 30 minutes in a dark humidity chamber at 37°C. After cover glass was mounted, dihydroethidium fluorescence signals were visualized using a fluorescence microscope (Observer D1; Zeiss, Germany). A total of 5 to 8 images from each sample were captured at fixed exposure times for all groups, and the fluorescence intensity per myocardial area was blindly quantified using AxioVision software.

Detailed Pharmacological Protocol for Vascular Studies

These solutions were kept at −20 °C and appropriate dilutions were made on the day of the experiment.

Gastroprotective Effects of C. procera
