sTRAIL Quantification in Serum

Quantifying Secreted TRAIL in Transduced MSCs

The culture supernatant of MSC-TRAIL was analyzed using ELISA according to manufacturer's procedure (Human TRAIL/TNFSF10 Quantikine ELISA Kit; R&D Systems, Inc.; cat. no. DTRL00). To investigate the persistence of transgene expression in vitro, BM-MSCs were seeded at a high density (4×104 per well of a 24-well plate). BM-MSCs were transduced with 50 multiplicities of infection of Ad-TRAIL in OPTI-MEM serum-free medium for 3 h in a 5% CO2 humidified 37°C chamber. The virus-containing medium was removed and additionally incubated in low-serum medium (Eagle's DMEM containing 2% FBS) at 37°C for ~40 days. Culture supernatants were harvested and fresh medium was changed every 3 day. Then, secreted TRAIL was examined.

Quantification of sfTRAIL Protein by ELISA

Quantifying TNFα and TRAIL Transcripts and Proteins

TNFα and TRAIL mRNA levels were determined by quantitative real-time PCR (qRT-PCR) analysis. Total RNA extraction and cDNA synthesis were performed as previously described [28 (link)]. The relative expression of the target gene transcript and reference gene transcript was calculated as ΔΔCt. 28S rRNA was used as reference gene. TRAIL protein levels in whole cell lysates were determined by Human TRAIL/TNFSF10 Quantikine ELISA Kit obtained from R&D Systems (Wiesbaden, Germany) and TNFα levels in the supernatant of cells were determined by Human TNFα Ultrasensitive ELISA Kit purchased from Invitrogen (Carlsbad, USA) according to the manufacturer's instructions.
