Synthesis and Characterization of Polymeric Nanoparticles

Moisture and air-sensitive materials were manipulated under nitrogen using Schlenk techniques. The chemicals anhydrous toluene (99.8%), hexane (≥95%), stannous octoate (92.5–100.0%) (SnOct2), methanol (99.8%), dimethylformamide (≥99.8%) (DMF), chloroform (≥99.5%), tetrahydrofuran (≥99.9%) (THF), Nile Red (≥97.0%) (NR), quercetin (≥95%) (Q), and 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (≥97.5%) (MTT) were acquired from Merck and used as received. The ε-caprolactone (97%) (ε-CL), acquired from Merck, was distilled in vacuo from CaH2 prior to use. Polyethylene glycol (PEG-2000 MW), acquired from Merck, was dried in vacuo over phosphorus pentoxide (P2O5) for 72 h.

For cell culture, human cervical cancer KB cells were obtained from the Korean Cell Line Bank (KCLB, Seoul, South Korea). RPMI 1640 medium, fetal bovine serum (FBS), penicillin, and streptomycin were purchased from Welgene (Seoul, South Korea). Cell Counting Kit-8 (CCK-8) was obtained from Dojindo Molecular Technologies (Tokyo, Japan). PEG-PLL-PLA triblock copolyelectrolyte was prepared as described previously.17 (link)

Fabrication of PLCL Sheet-form Scaffolds

PLCL was synthesized and subjected to a gel-pressing method to fabricate sheet-form scaffolds, as described elsewhere [25 (link)]. Briefly, PLCL (50:50) was synthesized at 150 °C for 24 h in the presence of Stannous Octoate (1 mmol, Sigma, St. Louis, MO, USA) as a catalyst. Dissolved PLCL (5% w/v, in Chloroform) was mixed homogeneously with approximately 300 μm sized salts to form the PLCL gels. The residual solvents were removed by placing the samples under vacuum for an additional 24 h. The salts were then leached out by immersing the samples in distilled, deionized water and subjecting them to constant shaking for 3 days. The resulting scaffolds (disk type, diameter = 9 mm and thickness = 3 mm) were freeze-dried for 24 h, and then sterilized with ethylene oxide gas. The porosity of the scaffolds was measured by a mercury intrusion porosimeter (Auropore IV; MicroMeritics, Norcross, GA, USA).

(St Louis, MO, USA) and used as received. 2-Methacryloyloxyethyl phosphorylcholine (MPC) was supplied by Nanjing Joy-Nature Ltd (Nanjing, China) and used as received. Doxorubicin (DOX)·HCl was neutralized with TEA to remove the hydrochloride to make it become hydrophobic DOX. Toluene and tetrahydrofuran (THF) (ChengDu KeLong Chemical Co., Ltd) were dried by refluxing over sodium and distilled before used. All the other reagents and solvents used were analytically pure and without further purification.

Synthesis and Characterization of Trimethylene Carbonate-Based Adhesives

Trimethylene carbonate adhesives were synthesized in a two-step reaction as described and validated previously (Bochyńska et al., 2013 ). Briefly, ring-opening polymerization was performed on TMC initiated by PEG with Sn(Oct)2 as a catalyst. The molar ratios, 4:1 and 6:1 for TMC2 and TMC3, resulted in two and three TMC moieties on each end of the PEG molecule, respectively (Figure 1). Oligomers were added dropwise to a volume of HDI under a flow of nitrogen to ensure an excess of HDI and an inert environment. Excess HDI was removed by precipitation in dry diethyl ether and the PEG400-(TMC3-HDI)2 was dried in a vacuum.
The chemicals were purchased from the following vendors: TMC from ForYou (China); HDI from Merck Schuchardt (Germany); diethyl ether from Biosolve (the Netherlands); PBS from B Braun Melsungen AG (Germany); and PEG (Mw = 400 g mol−1), acetic anhydride, stannous octoate (tin 2-ethylhexanoate, SnOct2) and chloroform-d (CDCl3) from Sigma Aldrich (the Netherlands). PEG was dried at 120 °C under vacuum, and diethyl ether was dried over molecular sieves prior to use.

HCAECs were acquired from Cambrex (Walkersville, MD, USA) and EGM-2 media with an MV bullet kit (Lonza, Basel, Switzerland). A cell-counting kit (CCK-8) was obtained from Dongin LS (Seoul, Korea). Phosphate-buffered saline (PBS) solution was supported from Hyclone (GE Healthcare Life Sciences, Boston, MA, USA). A Micro BCATM Protein assay kit was acquired from Thermo Scientific TM (Waltham, MA, USA). All chemicals were laboratory reagent grade and used without purification.
