Electrochemical Synthesis of Drug-Loaded Polypyrrole Films

Drug containing PPy films were grown potentiostatically onto the device electrodes using a Bio-Logic SP-150 potentiostat. The dexamethasone electrosynthesis solution consisted of 0.2 M Pyrrole (Alfa Aesar, Ward Hill, MA) and 0.02 M dexamethasone 21 phosphate disodium salt (Sigma-Aldrich, used as received) in Milli-Q water of 18 MΩ/cm resistivity. The Indomethacine electrosynthesis solution consisted of 0.2 M Pyrrole and 0.01 M Indomethacin (Sigma-Aldrich) in acetonitrile. The acetylsalicylic acid electrosynthesis solution consisted of 0.2 M Pyrrole (Alfa Aesar, Ward Hill, MA) and 0.02 M acetylsalicylic acid (Sigma-Aldrich,) in Milli-Q water. The films were synthesized by applying a constant current of 0.1 mA for Indomethacin and 1 mA for acetylsalicylic acid and dexamethasone each for 10 minutes between the drug release electrode and the counter electrode. For drug release experiments, the counter and working electrodes were reversed. The amount of released DEX, ASA and Indomethacin was quantified by absorbance at 242 nm, 298 nm and 318 nm, respectively using an infinite M200 Pro plate reader (TECAN).

Electrochemical Synthesis and Drug Release of PPy/DEX Films

Electrochemical Detection of SARS-CoV-2 Spike Protein

All chemicals were used as received without further purification. The chemicals were purchased as follows: pyrrole 98% − from Alfa Aesar (Germany), sulfuric acid (H2SO4; 96%) from Lachner (Czech Republic), nitric acid (HNO3, 63%), sodium hydroxide (NaOH, 98%), and chloroplatinic acid (H2PtCl6, 40% Pt) from Merck (Germany), potassium phosphate (KH2PO4, 98%) from Honeywell Riedel-de Haen (Germany), sodium chloride (NaCl, 99,5%), potassium chloride (KCl, 99,5%), and disodium hydrogen phosphate (Na2HPO4, 99%) from Carl Roth (Germany). SARS-CoV-2 spike glycoproteins were purchased from UAB Baltymas (Lithuania).
The experiment was performed using potentiostat/galvanostat Metrohm-Autolab model μAutolabIII/FRA2 μ3AUT71079 controlled by NOVA 2.1.3 software (EcoChemie, The Netherlands). All measurements were done in a homemade cell. The total volume of the cell was 250 μL. Three-electrode system consisted of a working electrode (WE) – Pt disk with 1 mm diameter sealed in glass, reference electrode (RE) – Ag/AgCl, and counter electrode (CE) – Pt disk of 2 mm diameter.

Oxidative Pulp Treatment Synthesis

Acrylic acid (AA, 98%) and iron (III) chloride hexahydrate (FeCl3·6H2O, 99%), were purchased from ACROS Organics (Geel, Belgium). 2,2,6,6-tetramethylpiperidine 1-oxyl (TEMPO, 98%+), 2-acrylamido-2-methylpropane sulfonic acid (AMPS, 98%), N,N′-methylenebisacrylamide (MBAA, 97%), pyrrole (Py, 98%), N,N,N’,N’-tetramethylethylenediamine (TMEDA, 99%), and ethylenediaminetetraacetic acid (EDTA, 99%) were used as received from Alfa Aesar (MA, US) without further purification.

Electrochemical Sensor Fabrication Protocol

4-Nitrobenzaldehyde (99%), tin(ii) chloride dihydride (SnCl2·2H2O, 98+%) were purchased from Acros Organics, terephthaloyl chloride (99%) and Nafion solution (5% w/w in H2O and 1-propanol) were obtained in Alfa Aesar, pyrrole (98%), pyridine (99%), sulfuric acid (H2SO4), potassium hydroxide (KOH) and 1.0 M PBS buffer (pH = 7) were purchased from Fisher Scientific and used without further purification. Carbon paper substrate was purchased from Fuel Cell Store (AvCarb P75T −40 × 40 cm), which is carbon particle-filled, polyacrylonitrile (PAN) based carbon fiber paper with hydrophobic Teflon wet-proof (13% wt treatment) coating nominal thickness of 0.255 mm (@ 1 psi/0.7 N cm−2).

cells), and QSG-7701 (human normal hepatocyte) cells were purchased
from the Shanghai Institute for Biological Sciences, Chinese Academy
of Sciences (Shanghai, China).
