PLGA Nanoparticles for β-Glucosidase Delivery

PLGA (Poly(D,L-lactide-co-glycolide), RG503H, MW ≅ 11,000) was purchased from (Evonik, Essen, Germany) and used as received. β-Glucosidase (β-Glu, MW 135 KDa), Tween® 20, 60, 80 (100%, guaranteed less than 3% water), and poly(vinyl alcohol) (PVA, MW 15,000), bovine serum albumin (BSA, 66 KDa, >98% pure), dichloromethane (DCM), 4-methylumbrelliferyl-β-D-glucopyranoside, and 4-methylumbelliferone (4-MU) were purchased from Sigma-Aldrich (Milan, Italy). MilliQ water was purified by a Millipore system (Millipore, Bedford, MA, USA). Analytical grade reagents were used for all other purposes unless otherwise noted.

Quantitative PLGA Composition Analysis

Poly(lactic-co-glycolic acid) with a GA/LA ratio of 50:50 (RG 503H) and 75:25 (RG 752H) was purchased from Evonik Nutrition & Care GmbH (Darmstadt, Germany). The reference standards for GA (99%) and LA (≥95%) were purchased from Sigma Aldrich (Darmstadt, Germany). Poly(D,L-lactic acid) (≥95%) was purchased from Polysciences, Inc. (Hirschberg an der Bergstraße, Germany). For the derivatization, 2,4′-dibromoacetophenone (≥98%) and triethanolamine (TEA) (≥98%) were purchased from Sigma Aldrich (Darmstadt, Germany). The solvents acetonitrile, acetone, tetrahydrofuran, ethyl acetate, dimethylformamide, and chloroform were all purchased from Sigma Aldrich (Darmstadt, Germany). For the hydrolysis and pH adjustment, potassium hydroxide (KOH) and aqueous hydrobromic acid (HBr) were purchased from Sigma Aldrich (Darmstadt, Germany). The pH indicator bromocresol green was purchased from Honeywell (Seelze, Germany).
