Biomolecular Analytical Techniques

All oligonucleotides were ordered from Integrated DNA Technologies (IDT), purified with HPLC and dissolved in PCR quality water (Invitrogen). DNA concentrations were measured using a NanoDrop 2000 spectrophotometer (Thermo Scientific). Diethylthiatricarbocyanine iodide (Cy7), dehydroisoandrosterone-3-sulfate (DIS) sodium salt dihydrate, deoxycorticosterone-21 glucoside (DOG) and all other chemicals were purchased from Sigma-Aldrich unless otherwise noted. Drug standards, including 3,4-methylenedioxypyrovalerone (MDPV), mephedrone, methylone, naphyrone and pentylone were purchased from Cayman Chemicals. Tween 20 and formamide were purchased from Fisher Scientific. Streptavidin-coated agarose resin (binding capacity: 1−3 mg biotinylated BSA/ml resin), One Shot Chemically Competent Escherichia coli, and SYBR Gold were purchased from Thermo Scientific. 500 μl micro-gravity columns were purchased from BioRad.

Radiolabeled Compounds and Drug Compounds Protocol

[3H]Methyl-4-phenylpyridinium ([3H]MPP+; 80 Ci/mmol) was purchased from American Radiolabeled Chemicals (St. Louis, MO, USA), while [3H]neurotransmitters (30–50 Ci/mmol) were purchased from Perkin Elmer (Shelton, CT, USA). Methylone, butylone, dibutylone, pentylone, and eutylone were purchased as hydrochloride salts from Cayman Chemical Company (Ann Arbor, MI, USA). All other chemicals and reagents were procured from MilliporeSigma (St. Louis, MO, USA) unless otherwise specified. Cocaine hydrochloride was obtained from the Pharmacy at the National Institute on Drug Abuse (NIDA) Intramural Research Program (IRP). Drugs for the in vivo experiments were dissolved in sterile saline and doses are expressed as the salts.

Quantitative Analysis of N-Ethylnorpentylone

N-Ethylnorpentylone reference material was purchased from Cayman Chemical (Ann Arbor, MI, USA) and MDMA-d5, used as internal standard (IS), was purchased from Cerilliant (Round Rock, TX, USA). Methanol and formic acid (98-100%) were purchased from Scharlau (Barcelona, Spain). Ammonium formate and tert-butyl methyl ether (MTBE) were purchased from Sigma-Aldrich (St Louis, MO, USA) and sodium tetraborate from LS Chemicals (Ribeirao Preto, SP, Brazil). Ultrapure deionized water was purified by a Milli-Q RG unit from Millipore (Billerica, MA, USA). All solvents employed in the sample extractions were HPLC grade. N-Ethylnorpentylone, pentylone and methylone used for monoamine transporter assays were obtained from Cayman Chemical. [3H]Norepinephrine (60 Ci/mmol) and [3H]methyl-4-phenylpyridinium ([3H]MPP+; 80 Ci/mmol) were purchased from American Radiolabeled Chemicals (St. Louis, MO, USA) while [3H]dopamine (28 Ci/mmol) and [3H]5-HT (38 Ci/mmol) were purchased from Perkin Elmer (Billerica, MA, USA). All other reagents used for monoamine transporter assays were obtained from Sigma-Aldrich unless otherwise indicated.
