Scfflox and Scfgfp were generated in a previous report (Ding et al. 2012 (link)). K14Cre (Dassule et al. 2000 (link)), K14CreERT (Vasioukhin et al. 1999 (link)), TyrCreERT2 (Bosenberg et al. 2006 (link)), R26-rtTA (Belteki et al. 2005 (link)), and tetO-DTA (Lee et al. 1998 (link)) mice were purchased from the Jackson Laboratory. K14-Scf (Kunisada et al. 1998a (link)) was kindly provided by Dr. John Harris (University of Massachusetts Medical School). Krox20Cre (Voiculescu et al. 2000 (link)), DhhCre (Skucas et al. 2013 (link)), PLPCreERT2 (Leone et al. 2003 (link)), CMVCreERT (Hayashi and McMahon 2002 (link)), R26-LacZ, and R26-YFP were described previously (Le et al. 2009 (link); Chen et al. 2014 (link)).

Genetic Mouse Models for Developmental Studies

All mice were housed and cared for according to MSSM and IACUC approved protocols. Ezh1deleted and Ezh2flox mice were previously reported [45 (link)]. EEDflox mice were provided by Weipeng Mu and Terry Magnuson [70 (link)]. Tcf/Lef:H2B-GFP mice were provided by Anna-Katerina Hadjantonakis [36 (link)]. Gli1LacZ, Ctnnb1flox, Smoflox, ShhEGFPCre, Atoh1-GFP, R26-rtTA, and Krt14-Cre mice were obtained from The Jackson Laboratory. Wild type C57BL/6 mice were obtained from Charles River Laboratories. Mice were genotyped by PCR using DNA extracted from tail skin. BrdU was administered as previously reported [51 (link)]. Briefly, BrdU was administered (50μg BrdU per 1g mouse weight) to mice or pregnant females 3–5 h before sacrificing [45 (link)].

Genetically Modified Mouse Strains

R26-floxed-rtTA, R26-floxed-GFP, R26-rtTA, CAG-rtTA, Ngn3-Cre, and NSG mouse strains were obtained from The Jackson Laboratory. Ngn3-GFP (Gu et al., 2002 (link); Jenny et al., 2002 (link); Lee et al., 2002 (link)) and Cdx2fl/fl (Silberg et al., 2002 (link); Verzi et al., 2013 (link)) mice have been described previously. The TetO-NPMcherry mouse lines were made by standard pronuclear injection at the Harvard Genomic Modification Facility (Cambridge, MA). All animal experiments are approved by the Harvard Institutional Animal Care and Use Committee.
