Rt2 microrna first strand kit

Manufactured by Qiagen

The RT2 microRNA First Strand Kit is a laboratory tool designed for the reverse transcription of microRNA molecules. It provides a standardized and efficient method for the conversion of microRNA into complementary DNA (cDNA) that can be used in subsequent qPCR or other molecular analysis applications.

Comprehensive Mouse microRNA Profiling

Check if the same lab product or an alternative is used in the 5 most similar protocols
For microRNA analysis, total RNA, including microRNA, was extracted from cell lysates using a miRNeasy Mini Kit (Qiagen cat#217004). Next, the RT2 microRNA First Strand Kit (Qiagen cat# 331401) was used to synthesize cDNA from 4 µg of total RNA. The microRNA PCR array (Mouse miRNome RT2 Whole Genome microRNA PCR Array, Qiagen cat#MAM-200A-2) was used to probe for 440 mouse microRNAs normalized to the average levels of the housekeeping small nuclear RNAs, SNORD 68, 66 and Rnu6.

Inflammatory microRNA Expression in Macrophages

Check if the same lab product or an alternative is used in the 5 most similar protocols
To determine the expression of inflammatory microRNAs, WT and 5-LO−/− macrophages were treated with or without LTB4 for 24 h. RNA was then extracted from the cells using a Qiagen RNeasy mini kit according to the manufacturer’s instructions. cDNA was synthesized using the RT2 microRNA First Strand Kit (Qiagen) and applied to PCR focused immunopathology microRNA array plates (Qiagen). Plates were processed in a BioRad CFX96 Connect Real-Time PCR System using automated baseline and threshold cycle detection.

Profiling Inflammatory microRNAs in Macrophages

Check if the same lab product or an alternative is used in the 5 most similar protocols
To determine the expression of inflammatory microRNAs, macrophages from C57BL/6 WT mice were treated with PTEN siRNA or scrambled siRNA as above. RNA was then extracted from the cells using Qiagen RNeasy mini kit according to the manufacturer’s instructions. cDNA was synthesized using the RT2 microRNA First Strand Kit (Qiagen) and applied to PCR focused immunopathology microRNA array plates (Qiagen) as previously shown (10 (link)). Normalization and statistical analysis of miRNA expression was conducted using SABiosciences’ Online PCR Array Data Analysis Web Portal as previously described (10 (link)).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
