MTT Assay for Cell Viability

Check if the same lab product or an alternative is used in the 5 most similar protocols
MTT assay measured the cell viability [20 (link)]. Briefly, 5 × 104 HaCaT or B16F10 cells cultured in a 24-well plate were exposed to UVA radiation using UVIlink CL-508L (UVItec, Cambridge, UK) or incubated with different test samples or inducers for the suggested time, with or without UVA irradiation. After PBS wash, cells were further incubated for 2 h in the presence of MTT (0.5 mg/mL). The cytoplasmic formazan crystals were solubilized with DMSO (0.8 mL). The intensity of color developed was estimated using an ELISA plate reader at 570 nm (λ570) (Bio-Tek Instruments, Winooski, VT, USA). Data were calculated from triplicates and denoted as cell viability percentage compared to unstimulated cells. The cell viability percentage of control/unstimulated cells was assigned as 100%.

In Vitro Cytotoxicity Evaluation of DSF/SPC-NSps

Check if the same lab product or an alternative is used in the 5 most similar protocols
In vitro cell cytotoxicity of DSF/SPC-NSps was evaluated via the MTT assay. 4T1 cells (1.0 × 104 cells/well in 150 μL) were sowed in 96-well plates and incubated 24 h in 5% CO2 at 37 °C. DSF/SPC-NSps with different concentrations and free DSF solution (DSF in DMSO) were added and incubated for 48 h. Then 20 μL of the MTT solution (5 mg/mL in PBS) were added into each well and incubated for 4 h. The medium was removed and 150 μL of DMSO was added to dissolve the formazan crystals. The maximum absorbance was measured by an ELISA plate reader at 570 nm (Biotek, USA). The cell viability rate was calculated as follows: Cell viability rate%=ODt/ODc100% (ODt: the mean optical density (OD) of the tested group; ODc: the mean OD of the control group. Each group’s half-inhibitory concentration (IC50) value was calculated by GraphPad Prism software, Version 7 (GraphPad Software, Inc., USA).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
