Morphine, Morphine-d3, tramadol, 13C-tramadol-d3, buprenorphine-d4, chlorpromazine-d3, and phenylbutazone were obtained as solutions from Sigma-Aldrich Chemie GmbH (Buchs, Switzerland). Butorphanol and buprenorphine were obtained as solutions from ReseaChem GmbH (Burgdorf, Switzerland) and Lipomed AG (Arlesheim, Switzerland), respectively. Ketoprofen-d3 and phenylbutazone-(diphenyl13C12) were purchased as powders Sigma-Aldrich Chemie GmbH (Buchs, Switzerland). Detomidine, acepromazine, meloxicam, ketoprofen, chlorpromazine, flunixin and fluphenazine were analytical standards available as powders in the authors’ laboratory. All solvents and other chemicals were purchased from Sigma-Aldrich Chemie GmbH (Buchs, Switzerland). All standards were dissolved in methanol, except for meloxicam that was dissolved in dimethylformamide, to yield stocks of 1 mg/ml. Firocoxib was obtained as an injectable for veterinary use. Chromatography-grade water used for LC-MS/MS analysis was processed by a PURELAB Option-Q system by ELGA LabWater (Labtec Services AG, Villmergen, Switzerland).

Quantification of Opiate Alkaloids in Poppy Seeds

Representative images of poppy seeds and the chemical structures of five OAs are shown in Figure 1 and Figure 2. Morphine, codeine, thebaine, noscapine, papaverine, Morphine-d3, and codeine-d3 were purchased from Sigma (St. Louis, MO, USA). The reference compounds were more than 99% pure, with identity and purity confirmed by chromatographic and spectral data (HRMS). Quantification of Morphine and Metabolites

All the standard molecules used in the study were purchased from Sigma-Aldrich (St. Louis, MO, USA): morphine, morphine-3-glucuronide, morphine-6-glucoronide solutions (1.0 mg/mL in methanol each standard) and 6-acetylmorphine (1.0 mg/mL in acetonitrile standard solution). Deuterated internal standards (ISs) were purchased from Sigma-Aldrich as well: morphine-D3 (1.0 mg/mL in methanol standard solution), 6-acethylmorphine-D3 (1.0 mg/mL in acetonitrile), morphine-3-glucuronide-D3 (100 µg/mL in methanol with 0.05% of NaOH) and morphine-6-glucuronide-D3 (100 µg/mL methanol/water (1:1, v/v)). Solvents used in the extractive processes were purchased from Sigma-Aldrich (methanol, hydrochloric acid, chloroform, dichloromethane and ethyl acetate) and VWR Chemicals (Radnor, PA, USA) (acetone, acetonitrile, isopropanol and n-hexane). Buffer pH 6.88 solution was purchased from PanReac (Barcelona, Spain). For histological inclusions, absolute ethanol and xylene substitute were purchased from VWR Chemicals and high fusion point paraffin pearls Histowax® were purchased from International Medical Products (Brussel, Belgium).

Quantitative Analysis of Opioids

Morphine, methadone, codeine, 6-MAM, EDDP standards, and Morphine-d3 and methadone-d3 internal standards (IS) were purchased from Sigma Aldrich (Milan, Italy). For the extraction procedure, distilled water (H2O) for analysis, sodium hydroxide (NaOH), methanol (MeOH) for analysis (≥99.9%), hydrochloric acid for analysis (37%), dichloromethane for analysis stabilized with amylin (≥99.9%), 2-propanol (99.8%) and ammonium hydroxide (32%) of reagent grade were purchased from Carlo Erba (Cornaredo, Italy). For the UHPLC analysis, methanol for UHPLC-MS (99.99%), ultrapure water for UHPLC-MS and formic acid (FA) for LC-MS (≥99%) were purchased from Carlo Erba (Cornaredo, Italy). All reagents and laboratory consumables were stored according to the manufacturer’s instructions.

Quantification of Opioids in Urine
