Cell Culture of Cancer and Endothelial Cell Lines

Check if the same lab product or an alternative is used in the 5 most similar protocols
HT-29 colon cancer cell line and 2H-11 mouse endothelial cell lines were purchased from the American Type Culture Collection (ATCC; Manassas, VA, USA), while HUVEC was purchased from Lonza (Basel, Switzerland). Cells were maintained in a complete medium, either Dulbecco’s modified Eagle’s medium (DMEM; Life Technologies, Carlsbad, CA, USA) containing 10% heat-inactivated fetal bovine serum (FBS) (Corning, NY, USA), Penicillin (100 U/mL), Streptomycin (100 µg/mL) (Life Technologies) and 2 mM L-alanyl-L-glutamine dipeptide (GlutaMAX Supplement; Life Technologies) for HT-29 and 2H-11 or EBM™-2 Endothelial Cell Growth Basel Medium, supplemented with EBM™-2 Endothelial SingleQuots™ kit (Lonza) for HUVEC. Cells were grown under standard culture conditions (37 °C with 5% CO2) and passaged no more than 5 times for HUVEC and no more than 20 times for HT-29 and 2H-11. All cell lines were mycoplasma-free, as determined using the MycoAlert™ Mycoplasma Detection Kit (Lonza) and/or a custom PCR-based assay, as described previously [11 (link),51 (link)].

Maintenance of Colon Cancer Cell Lines

Check if the same lab product or an alternative is used in the 5 most similar protocols
An HT29 colon cancer cell line was purchased from American Type Culture Collection (Manassas, VA, USA), and SW480 and SW620 colon cancer cell lines were purchased from the Bioresource Collection and Research Center (Hsinchu, Taiwan). All cell lines were maintained in Dulbecco’s Modified Eagle Medium (Gibco, Grand Island, NY, USA) supplemented with 4.5 g/L D-glucose, 10% fetal bovine serum (Gibco), and 1% penicillin/streptomycin (Gibco) in a 37 °C incubator with 5% CO2.

Culturing Colon Cancer and Normal Fibroblast Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
The HT29 colon cancer cell line was purchased from the American Type Culture Collection (Manassas, VA, USA) and the CCD-18Co normal colon fibroblast cell line was purchased from the Korean Cell Line Bank (Seoul, Korea). The colon cancer cell line was cultured in Dulbecco’s modified Eagle’s medium (DMEM; HyClone, Logan, UT, USA) and the CCD18-Co cell line was cultured in RPMI-1640 medium (WelGENE, Daegu, Korea) supplemented with 10% heat-inactivated fetal bovine serum (HyClone) at 37°C in a 5% CO2 incubator.

Antiproliferative Activity of 3-Phenylthiazolo[3,2-a]benzimidazoles

Check if the same lab product or an alternative is used in the 5 most similar protocols
Anti-proliferative activity of the synthesised 3-phenylthiazolo[3,2-a]benzimidazoles 4a–d was evaluated at Stem Cell Therapy and Tissue Reengineering Program, King Faisal Specialized Hospital and Research Center, Riyadh, Saudi Arabia. In vitro anti-proliferative activity was measured by the cell growth inhibition assay. This assay was conducted by use WST-1(water soluble tetrazolium-1) reagent26 for determination of IC50 for each compound. HT-29 colon cancer cell line and MDA-MB-468 triple negative breast cancer cell line were purchased from the American Type Culture Collection. Cells were maintained in RPMI 1640 (Sigma-Aldrich, St. Louis, MO), supplemented with 10% FBS (Fetal Bovine Serum) (Lonza Group, Basel, Switzerland), 100 IU/mL penicillin, 100 mg/mL streptomycin, and 2 mmol/L L-glutamine (Sigma). Cells were seeded into 96-well plates at 0.4 * 104/well and incubated overnight. The medium was replaced with fresh one containing the desired concentrations of the test compounds. After 48 h, 10 μl of the WST-1 reagent were added to each well and the plates were re-incubated for 4 h at 37 °C. The amount of formazan was quantified using ELISA (Enzyme Linked Immunosorbent Assay) reader at 450 nm.

The human HT-29 colon cancer cell line was obtained from the American Type Culture Collection (Manassas, VA, USA). The cells were maintained in Dulbecco's modified Eagle's medium (4.5 g/l glucose), supplemented with 10% fetal calf serum, l-glutamine and antibiotics (Biological Industries, Beit Haemek, Israel) at 37°C with 5% CO2 in a humidified incubator.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
