Cell Culture of NK-92MI Cells

Culturing NKTCL and NK-92 MI Cell Lines

Three NKTCL cell lines—SNT-8, SNK-10, and SNT-16 (kind gifts from Dr. Norio Shimizu at Tokyo Medical and Dental University)—were cultured in RPMI-1640 medium (Hyclone) supplemented with 10% human plasma (Gemini) and 700 U/mL recombinant human interleukin-2 (PeproTech). The NK-92 MI cell line was purchased from ATCC (CRL-2408) and cultured in Alpha Minimum Essential medium without ribonucleosides and deoxyribonucleosides (Gibco) but with 2 mM L-glutamine and 1.5 g/L sodium bicarbonate (Life Technologies) supplemented with 12.5% fetal bovine serum (Biological Industries), 12.5% horse serum (Biological Industries), 0.2 mM inositol (Solarbio), 0.1 mM 2-mercaptoethanol (Solarbio) and 0.02 mM folic acid (Sigma Aldrich). All cells were cultivated at 37°C in a humidified atmosphere of 5% CO2 in the air.
