Authentication of PDAC cell lines

Cells passaged for less than 6 months were authenticated by DNA fingerprinting analysis using short-tandem repeat (STR) markers.

Pancreatic Cancer Cell Culture and IFN-γ Treatment

PANC-1, CFPAC-1, and BxPC-3 cells were obtained from China Infrastructure of Cell Line Resources. The cells were maintained in Dulbecco's modified Eagle's medium (Hyclone Co., Logan, UT, USA) supplemented with 10% fetal bovine serum (Hyclone Co., Logan, UT, USA) and 100 U/ml penicillin and streptomycin. The cells were grown at 37°C in a humidified 5% CO2 air incubator and subcultured twice a week. For treatment, the cells were washed twice with phosphate-buffered saline (PBS) and 1 × 106 cells were incubated in cell-culture plates with or without 500 U/ml interferon-γ (IFN-γ; Sigma Co., St. Louis, MO, USA) for 48 h.

Culturing and Maintaining Pancreatic Cancer Cell Lines

Human pancreatic cancer cell line PANC-1, SW1990, and BxPC-3 were obtained from China Infrastructure of Cell Line Resources (Beijing, China). Capan-2 and MIA PaCa-2 were purchased from Obio Technology (Shanghai, China) Corp., Ltd. SW1990, PANC-1 and MIA PaCa-2 were cultured in DMEM, Capan-2 was maintained in McCoy’s 5A and BxPC-3 was cultured in RMPI 1640, all supplemented with 10% fetal bovine serum, 100 U/ml of penicillin and 100 μg/ml of streptomycin. All of the cells were incubated at 37 ℃ in a humidified atmosphere of 5% CO2. The gemcitabine (Gemzar) drug was purchased from Eli Lilly and Company (Indianapolis, Indiana, USA).

Culturing PC Cell Lines in Mice

BALB/c male nude mice were purchased from Vital River Laboratory Animal Technology Co., Ltd. (Beijing, China). The mice experiments were approved by the Animal Experimental Ethics Committee of PLA General Hospital and conducted in the animal facility of PLA General Hospital according to the animal experimental ethics committee guidelines of PLA General Hospital.
