Microneedle Delivery of Parkinson's Drugs

Stainless steel microneedle rollers (500 μm) were purchased from Pearl Enterprises LLC. (Lakewood, NJ, USA) Amantadine hydrochloride, pramipexole dihydrochloride, and 0.1 M isotonic phosphate buffered saline (PBS) were purchased from Sigma–Aldrich Co. (St. Louis, MO, USA). Amantadine hydrochloride and pramipexole dihydrochloride were reconstituted using PBS. The water was processed using NanoPure Infinity Ultrapure water purification system (Barnstead, Dubuque, IA, USA).

Pharmacological Evaluation of Dopamine Agonists

Dopamine Receptor Modulator Preparation

DA hydrochloride (Lot# BCBG8676V), pramipexole dihydrochloride (Batch# 0000069160), (±)-7-Hydroxy-2-(di-n-propylamino) tetralin hydrobromide (7-OH-DPAT, Lot# 037M4153V), desipramine hydrochloride (Lot# 107K0730) and nomifensine maleate (Lot# 087K4064) were purchased from Sigma-Aldrich (St. Louis, MO). SB277011A dihydrochloride (Batch# 3B/219051) was purchased from Tocris Bioscience (Bristol, UK). Purified (18.2 MΩ) water was used to prepare all aqueous solutions. The artificial cerebrospinal fluid (aCSF) modified for zebrafish contained 131 mM NaCl, 2 mM KCl, 1.25 mM KH2PO4, 20 mM NaHCO3, 2 mM MgSO4, 10 mM glucose, 2.5 mM CaCl2·H2O, and 10 mM HEPES (pH 7.4). Perchloric acid (0.2 M) was used to prepare the DA stock solutions and aCSF without glucose was used to dilute the stock solutions for electrode calibrations. Stock solutions of 50 μM 7-OH-DPAT, 50 μM pramipexole, 10 μM nomifensine, and 10 μM SB277011A, were freshly prepared before each experiment. 7-OH-DPAT and SB277011A stock solutions were prepared in DMSO and nomifensine and pramipexole stock solutions were prepared in aCSF.

Dopamine Receptor Drugs Protocol

(±)-7-hydroxy-2-(di-n-propylamino)tetralin hydrobromide (7-OH-DPAT) was received from the National Institutes of Mental Health Chemical Synthesis and Drug Supply Program. Pramipexole dihydrochloride and S-(−)-Raclopride were purchased from Sigma-Aldrich (A1237; R121; St. Louis, MO). 7-OH-DPAT, pramipexole, and raclopride were dissolved in 0.9% sterile physiological saline and administered subcutaneously. SB-277011A was obtained from MEGAPharma Kft (Budapest, Hungary) and dissolved in 25% w/v hydroxypropyl-β-cyclodextrin in sterile water and administered intraperitoneally. All solutions were made fresh daily and administered in a volume of 1 ml/kg.

Aripiprazole was purchased from Santa Cruz Biotechnology (Dallas, TX). Forskolin was purchased from Tocris (Ellisville, MO). Haloperidol was a gift from D. Nichols, Purdue University.

Larval Behavioral Responses to Dopaminergic Drugs

The larvae were exposed to two different drugs during the behavioural recordings, pramipexole dihydrochloride and L-Dopa ethyl ester (both Sigma-Aldrich, St. Louis, USA, CAS: 104632-25-9 and 37178-37-3). Drug preparation was performed at the day of recording. Drugs were diluted using distilled water to reach their respective concentrations. For pramipexole and L-Dopa, larvae were submerged in 1 μM, 10 μM and 100 μM doses. Drugs were administered into the wells between 11:30 and 12:30 on the day of recording.
