Point and deletion mutants of SLFN5 were generated using the Stratagene QuickChange Site-Directed Mutagenesis protocol. pLKO.1-shSLFN5 plasmids (RHS4533-EG162394) were purchased from Dharmacon. pX330-GFP-Cas9 plasmid for human SLFN5 knockout was constructed with single guide RNA (5’-GATGCAGGAAAAGTCACCCT-3’).
