Multiparametric Flow Cytometry Analysis

The anti-human mAbs have been used: CD3 PE (from Beckman Coulter), CD3 PErCP, CD11b PE, CD14 APC, CD56 PE, CD56 APC, CD73 PE, CD107a FITC, Granzyme A FITC, Perforin FITC, pJNK PE (from BD Biosciences), HLA-DR FITC (Dako), CD44 FITC, CD105 FITC, Granzyme B PE, Granzyme K Alexa647 (Immunotools), CD90 APC, COX2 FITC (Cayman chemicals). Each flow cytometric analysis was controlled with isotype-matched mAbs. All flow cytometry-based experiments were performed on FACS Calibur using Cell-Quest Pro Software. Offline data analysis was done on Summit 5.1 software.

Immunophenotyping of Leukemia Cells

A variety of Ph+ leukemia cells and K562 cells carrying either Snail or Twist1 were re-suspended in PBS plus 0.5% BSA to a density of 1 × 107 cell/ml. Antibodies against CD34 PE (#IM1459u, Beckman Coulter), CD117 PCy7 (#IM3698, Beckman Coulter), CD45 KRO (#A96416, Beckman Coulter), CD61 PC7 (#IM3716, Beckman Coulter), CD41 FITC (#9011-0419, eBioscience), CD44 FITC (#F7083, DAKO, Denmark), CD44 FITC (#9011-0441, eBioscience) and CD184 APC (#FAB173A, R&D Systems, Minneapolis, MN, USA ) were titrated for use or added at the volumes suggested by the manufacturer and the mixture was incubated on ice for 40 min. After washing with PBS and 0.5% BSA, the labeled cells were analyzed on a Beckman Coulter Navios cytometer.

Phenotyping of Cultured hAM-MSCs

The expression of MSCs-specific surface markers was studied on the cultured hAM-MSCs after passage 3. The cells were separately stained with a set of fluorochrome-conjugated antibodies [CD29-FITC, CD105-FITC, CD44-FITC, CD73-PE, CD34-PE, CD166-PE and CD45-FITC (all from Dako, Produktionsvej, Denmark)] for 45 min at 4 °C. After the washing step, the cells were analyzed by flow cytometry technique (Partec 3, USA).
