Telemetric Arterial Blood Pressure Monitoring

The right femoral artery was isolated, and the catheter of the telemetric device was inserted into the right femoral artery. Through the same ventral incision, a pocket along the right flank was formed. The body of the telemetric device was placed in the pocket and secured with tissue adhesive (Vetbond, 3 M Animal Care Products, St Paul, MN, USA). The ventral incision was then closed with suture. All animals were allowed 1 week for recovery before being used in experiments.

Measuring Blood Pressure and Heart Rate in Rats

Telemetric Blood Pressure Monitoring in Marmosets

Marmosets were pre-medicated with ketamine (Ketavet; 10 mg/0.1 ml i.m. injection; Henry Schein, Melville, NY) before receiving the analgesic Carprofen (1.5 mg/0.03 ml s.c.; Pfizer, Kent, UK). They were intubated and maintained on 2.0–2.5% isoflurane in 0.3 l/min O2. Animals were monitored throughout using pulse-oximetry and capnography (Microcap Handheld Capnograph, Oridion Capnography Inc., MA, USA) as well as a rectal thermometer measuring core body temperature (TES-1319 K-type digital thermometer, TES Electrical Electronic Corp, Taipei, Taiwan). The descending aorta was visualized within the abdominal cavity and the probe catheter of a telemetric blood pressure transmitter (HDS-10, Data Sciences International, USA) was implanted into the aorta just above the aortic bifurcation. Blood flow was occluded for no more than 3 min. The probe was sutured in place within the abdomen. All monkeys received meloxicam (0.15 mg/0.1 ml p.o.; Boehringer Ingelheim) postoperatively for three days in addition to prophylactic antibiotic treatment with amoxicillin and clavulanic acid (Synulox; 50 mg/ml solution, Pfizer, Kent, UK) one day before and for six days after surgery.

Effects of Arhgef11 Deletion on Salt-Induced Albuminuria

At 4 weeks of age, age-matched male SS-WT (n=9) and SS-Arhgef11−/− (n=10) were weaned onto a low-salt diet (0.3% NaCl). Twenty-four-hour urine collections were done at 6 weeks of age on low salt and then each group was placed on high salt (2.0% NaCl) for the remainder of the study. Albuminuria was measured using a rat albumin ELISA kit (Abcam, ab108789). At week 7, a subset of animals from each group (n=8) were implanted with telemetry transmitters (model HD-S10, Data Sciences International) as previously described.13 (link),16 (link) After 1 week of recovery, blood pressure was measured for 24 hours one day each week from weeks 8 to 15, followed by 24-hour urine collections were made for the determination of proteinuria and albuminuria. Euthanasia was performed after week 15 blood pressure, and urine collections were complete. Organ weights, serum, and kidney samples for histological examination were collected.

Radiotelemeter Surgical Implantation in Rats

Under isoflurane anesthesia (1.5% in oxygen), a radiotelemeter transmitter (HD-S10; Data Sciences International, MN, USA) was implanted subcutaneously through a 1–1.5 cm incision in the left inguinal area. The radiotelemeter catheter tip was introduced into the left femoral artery and advanced into the abdominal aorta below the renal artery. During the same surgery, an additional femoral catheter (polyethylene 50 tubing; Instech Laboratories, PA, USA) was implanted into the left femoral vein and externalized between the scapulae. During surgery, all animals received a dose of enrofloxacin (Baytril®, 2.5 mg/kg, im) and carprofen (Rimadyl®, 5 mg/kg, sc). To allow rats to move freely, they were placed in a tether jacket and the external portion of the vein catheter was advanced into a protective spring which was attached to a swivel cage top.

Telemetric Monitoring of Rat SCI

All animal housing conditions, surgical procedures, and postoperative care were conducted according to the University of Kentucky Institutional Animal Care and Use Committee and the National Institutes of Health animal care guidelines. Animals were housed in a temperature and humidity-controlled room with a 12/12-hour light/dark cycle. Efforts were made to minimize unnecessary foot traffic and other potential environmental disturbances in the room. One week prior to SCI, a total of 24 naïve 3 to 3.5 month old female Wistar rats (250-275 grams) were anesthetized (2% isoflurane) and implanted with telemetric blood pressure transmitters (model HD-S10, Data Sciences International, Inc., St. Paul, MN) into the descending abdominal aorta according to the manufacturer’s surgical guidelines as previously reported (Eldahan et al., 2018 (link); Rabchevsky et al., 2012 (link)). Telemetry probes were secured intra-abdominally by suturing to the ventral abdominal wall using non-absorbable silk sutures before closing the skin with 3-0 vicryl sutures. Animals were treated post-operatively as described below.
