Peptide Purification and Analysis Reagents

Tyrothricin, bacitracin, propionic acid, sodium pentane sulfate, sodium sulfate and triethylamine were bought from Sigma-Aldrich (Diegem, Belgium), whereas vancomycin HCl was purchased from Bufa (Ijsselstein, the Netherlands). These peptides were ordered without explicit pharmacopoeial quality requirements, in order to increase the probability of containing related impurities. Bleomycin sulfate was obtained from Sanofi Aventis (Brussel, Belgium). Acetonitrile, methanol and tetrahydrofuran of HPLC grade were acquired from Fisher Scientific (Aalst, Belgium), whereas Acetonitrile, methanol and trifluoroacetic acid (TFA) of UPLC grade were bought from Biosolve (Valkenswaard, the Netherlands). Water was purified using an Arium 611 purification system (Sartorius, Gottingen, Germany) yielding≥18.2 MΩ×cm quality water. Potassium dihydrogen phosphate and ammonium sulfate were purchased from Merck (Overijse, Belgium), whereas dipotassium hydrogen phosphate was obtained from Panreac Chimica (Barcelona, Spain).

Antimicrobial Peptide Screening Assay

Gramicidin S, gramicidin D, tyrothricin, gentamicin, resazurin sodium salt, and KCl was supplied by Sigma (St. Louis, MO, USA). Acetonitrile, HPLC-grade far UV cut-off was supplied by Romil Ltd. (Cambridge, UK). Merck (Darmstadt, Germany) supplied agar, yeast extract, tryptone, Na2HPO4, and KH2PO4; and Merck (Wadeville, South Africa) supplied sodium chloride, brain heart infusion broth, and black 96-well polystyrene plates. From Corning (Kennebunk, ME, USA), 96-well polystyrene plates were acquired, and cellulose filters (Paper) (MN 615/No 1) were obtained from Macherey-Nagel (Düren, Germany). Mixed cellulose ester filters (HAWP, GSWP) were supplied by Waters-Millipore (Milford, MA, USA). Polycarbonate filters were supplied by Nuclepore Corp (Plesanton, CA, USA), and cellulose nitrate was provided by Sartorius (Gottingen, Germany). AcroPrepTM Advance 96 filter plates were sourced from Pall Corporation (Ann Arbor, MI, USA). Analytical grade water was obtained filtering water from a reverse osmosis plant through Millipore Milli Q® water purification system (Milford, MA, USA). Melittin, magainin 2, leucocin A, and PGLa was sourced from of the BIOPEPTM peptide library (Stellenbosch University, South Africa).
