Investigating ADORA2A and STAT3 in Mice

The ADORA2A-knockout (gKO) mice used in the study were obtained from Dr. Jiang-Fan Chen (Molecular Neuropharmacology Laboratory, Boston University School of Medicine, Boston, MA, USA). Wild-type (WT) C57 mice were purchased from the Department of Laboratory Animal Science, Army Medical University, China. All mice in this study were 8–9 weeks old and weighed 25–28 g. All animal experiments and care were approved by the Third Military Medical University Committee on Ethics in the Care and Use of Laboratory Animals.
The ADORA2A-specific agonist CGS21680 (Cat. No. 1063) and -specific inhibitor SCH58261 (Cat. No. 2270) were purchased from Tocris Bioscience (Abingdon, UK). Gefitinib-Resistant Lung Cancer Cell Lines

PC‐9 and HCC827 human lung adenocarcinoma cell lines and HEK293T human embryonic kidney cell line were purchased from the Cell Bank of the Chinese Academy of Sciences (Shanghai, China). Acquired gefitinib‐resistant PC‐9/GR and HCC827/GR cells were established by selecting parental cells with gradually elevated concentrations of gefitinib up to 2 μM for 2 months as described previously. Cells were maintained in RPMI 1640 medium (Hyclone) supplemented with 10% fetal bovine serum (Invitrogen) and 1% penicillin–streptomycin (Invitrogen) at 37°C in a humidified incubator containing 5% CO2.
The β‐elemene injection (National Medical Product Administration Approval Number: Chinese medicine H10960114) was obtained from Dalian Huali Jingang Pharmaceutical (Dalian, China). The STAT3‐specific activator colivelin (sc‐361 153) was purchased from Santa Cruz Biotechnology (TX, USA).
