Evaluating c-Kit-deficient Mouse Model

Experiments were done with 3-month-old c-Kit deficient mice (KitW/W-v) and littermate (Kit+/+) controls. KitW/W-v mice carry a c-Kit null mutation (W) and a hypomorphic allele (W-v). Compound heterozygotes exhibit reduced kinase activity and represent the most severe c-Kit deficient genotype that is not embryonic lethal [18 (link)]. KitW/W-v and Kit+/+ mice are obtained from the F1 generation of WB/ReJ KitW/J and C57BL/6J KitW-v/J crosses (Stock No. 000692 and 000049, Jackson Laboratories). Females and males were equally represented in all experimental groups. All experiments were conducted according to IACUC approvals and NIH guidelines.

All animal usage was approved by Cornell University’s Institutional Animal Care and Use Committee, under protocol 2004-0038 to J.C.S. Four- to eight-day-old KitW/Wv male mice generated by crossing C57BL/6J-KitWv/J and WB/ReJ KitW/J were used in this study. C57BL/6J-KitWv/J (#000049), WB/ReJ KitW/J (#000692) and B6(Cg)-Tyrc-2J/J (#000058) mice were purchased from The Jackson Laboratory. The Stella-eGFP transgenic ESC line (Stella-eGFP ESCs) with Stella flanking sequences coupled to eGFP as the reporter was kindly provided by Prof. M. Azim Surani 19 (link).
