Fluoxetine Affects Rodent Behavior

The selective serotonin reuptake inhibitor (SSRI) fluoxetine was purchased from Wako Pure Chemical Industries, Ltd. A fluoxetine dose of 10 mg/kg was dissolved in 0.2 ml of 0.9% saline and administered intraperitoneally (i.p.) approximately 10 minutes before tests. This dose of fluoxetine was selected due to reports from previous studies finding that fluoxetine administration at this dose causes several behavioral changes in rodents55 (link),56 (link). As a control treatment, the equivalent volume of saline (SAL) was given to animals.

Preparation of Pharmacological Agents

Decynium‐22 was purchased from Tocris Bioscience, corticosterone was purchased from Sigma‐Aldrich, and fluoxetine was purchased from FUJIFILM Wako Pure Chemical. Stock solution of decynium‐22 and corticosterone was prepared in DMSO at concentration of 1 mM and 10 mM, respectively. Stock solution of 10 mM fluoxetine was prepared in distilled water. These stock solutions were stored at −30℃ and were diluted in Ringer's solution freshly before the experiments.

Fluoxetine Exposure in Mice

Fluoxetine (0.8 mg/mL; Wako Pure Chemical Industries, Osaka, Japan) was supplied to group-housed male mice in drinking water from when the mice were 8-weeks-old until the end of the experiment. Fluoxetine-treated mice were subjected to the behavioural tests (n = 16 cages) described above and gene expression analysis of the hippocampus (n = 8) was performed as described above.
