Cortisol LFIA Strip Immunoassay

Cortisol, polyclonal anti-HRP antibody produced in rabbit, bovine serum albumin (BSA), and Tween-20 were purchased from Sigma Aldrich (St. Louis, MO). Western Lightning Ultra CL substrate for HRP was purchased from Perkin Elmer (Waltham, MA). Polyclonal antiCortisol antibody produced in rabbit was purchased from Analytical Antibodies (Bologna, Italy). HRP-conjugate Cortisol was generously provided by Diametra (Milano, Italy). Phosphate buffered saline (PBS) was prepared as follows: 10 mmol L -1 Na2HPO4, 2 mmol L -1 KH2PO4, 137 mmol L -1 NaCl, 2.7 mmol L -1 KCl, pH 7.4. The other reagents were of analytical grade and were employed as received.
Materials for the LFIA strip: glass fiber pad, cellulose pad, nitrocellulose membrane (Hi-flow plus 180 membrane cards), and cellulose adsorbent pad were from Merck Millipore (Billerica, MA); saliva sample pad I (Whatman LF1) and saliva sample pad II (Whatman Standard 14) were provided by GE Healthcare Life Science (UK).

Quantitative Multiplex Assay for Mycotoxins

The -globulin fraction was isolated by ammonium sulfate precipitation and used without any additional treatments. Jeffamine M1000 (1000 g/mol) was kindly provided by Huntsman (Belgium).
Millipore High Flow (HF) 240, 180, 120 and 90, and cellulose adsorbent pad were obtained from Merck Millipore (Billerica, MA, USA).

Optimized Lateral Flow Immunoassay for Ochratoxin A

The polyclonal anti-OTA antibody produced in rabbit and the HRP-OTA conjugate were kindly provided by Euroclone (Milan, Italy). Polyclonal anti-HRP antibody produced in rabbit, ovalbumin (OVA), Tween-20, and polyethylene glycol 10000 were obtained from Sigma-Aldrich Co (St. Louis, MO). The CL HRP detection substrate Supersignal ELISA Femto was purchased from Thermo Scientific Inc. (Rockford, IL). The other reagents were of analytical grade and were employed as received. Phosphate buffered saline (PBS) contained 10 mmol L -1 Na2HPO4, 2 mmol L -1 KH2PO4, 137 mmol L -1 NaCl, 2.7 mmol L -1 KCl, with pH adjusted to 7.4. The LFIA strips were produced using Whatman Standard 14 glass fibre sample pad (GE Healthcare Lifescience, Chalfont St. Giles, UK), Hi-flow plus 180 nitrocellulose membrane cards (Merck Millipore, Billerica, MA), and cellulose adsorbent pads (Merck Millipore).

Immunoassay with Rabbit Polyclonal Antibodies

Polyclonal anti-human Albumin antibody produced in rabbit, polyclonal anti-peroxidase (HRP) antibody produced in rabbit, HRP-labeled goat anti-rabbit immunoglobulin, human serum albumin and bovine serum albumin were purchased from Sigma-Aldrich (St. Louis, MO).
All other reagents were of analytical grade and were used as purchased.
The SuperSignal ELISA Femto CL substrate for HRP was obtained from Thermo Fisher Scientific, Inc. (Rockford, IL).
Nitrocellulose membranes (Hi-flow180 A4 sheet) and cellulose adsorbent pads (20 x 300 mm) were bought from Merckmillipore (Billerica, MA). Sample pads were Standard 14 glass fiber pads (22 x 300 mmm) from Whatman and were purchased from GE Healthcare Life Sciences (Buckinghamshire, UK).
Phosphate buffered saline (PBS) was prepared as follows: 10 mmol L -1 Na2HPO4, 2 mmol L -1 KH2PO4, 137 mmol L -1 NaCl, 2.7 mmol L -1 KCl (pH 7.4).
Synthetic urine was prepared as previously described [36] .
