Phagocytosis of Labeled Tumor Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
Imaging was performed on an Applied Precision DeltaVision (GE healthcare) deconvolution microscope. The phagocytosis assay was prepared as stated above with 3×105 macrophages per well in an 8 well Lab-Tek Chambered Coverglass (Thermo Scientific). 1×105 CFSE labeled B16F10 cells were incubated with the proteins and then added to the macrophages for at least 3 hours before imaging. Samples were stained with anti-human CD14 Alexa Fluor 647 antibody before imaging on the DeltaVision microscope. Images were recorded on the 20× lens and 60× oil lens.

Mitochondrial Membrane Potential in Fibroblasts

Check if the same lab product or an alternative is used in the 5 most similar protocols
The mitochondrial membrane potential was assessed in live primary fibroblasts using 5,5′,6,6′-tetrachloro-1,1′,3,3′-tetraethylbenzimidazolylcarbocyanine iodide (JC-1; Sigma-Aldrich) as described [26 (link)]. Briefly, fibroblasts were washed and incubated with 5 µg/ml JC-1 dye for 20 min at 37℃. The cells were then rinsed with culture medium, and their images were obtained using the Applied Precision DeltaVision fluorescence microscopy system (GE Healthcare, Chicago, IL, USA). JC-1 accumulates as red fluorescent aggregates within polarized mitochondria but does as green fluorescent monomers within less polarized mitochondria.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
