Xanthine oxidase fluorometric assay kit

Manufactured by Cayman Chemical
Sourced in United States

The Xanthine Oxidase Fluorometric Assay Kit is a tool used to quantify the activity of the enzyme xanthine oxidase. It utilizes a fluorometric detection method to measure the oxidation of a substrate, providing a sensitive and quantitative assessment of xanthine oxidase levels.

Hypoxia-Induced Oxidative Stress Evaluation

Check if the same lab product or an alternative is used in the 5 most similar protocols
Hypoxia induced oxidative stress was evaluated using enzymatic and non-enzymatic cellular anti-oxidants i.e. free radical generation by ROS estimation, GSH/GSSH, lipid peroxidation and xanthine oxidase assays at different time points of 1, 3, 6, 12 and 24 h of hypoxia by quantitative methods.
Free radical generation was assessed by the method described by Cathcart et. al. [48 (link)]. Reduced glutathione (GSH) and oxidised glutathione (GSSG) levels in cytosolic fractions were determined fluorimetrically by the method of Hissin and Hilf [49 (link)]. Lipid peroxidation was measured in cellular cytosolic fraction in terms of formation of malondialdehyde as described by Utley et. al. [50 (link)]. To evaluate generation of superoxide radicals under hypoxia, assessment of xanthine oxidase activity was done in cytosolic fractions of HVCM cells using Xanthine Oxidase Fluorometric assay kit (Cayman Chemical, 10010895) according to manufacturer’s instructions.

Liver Xanthine Oxidase Measurement

Check if the same lab product or an alternative is used in the 5 most similar protocols
After the livers were homogenized and centrifuged, the supernatant was collected. The xanthine oxidase activities were detected using a xanthine oxidase fluorometric assay kit (Cayman Chemical, Ann Arbor, MI, USA). The procedures were followed according to the manufacturer’s instructions.

Inflammatory Biomarkers Quantification

Check if the same lab product or an alternative is used in the 5 most similar protocols
ELISA kits were used to measure the serum and kidney inflammatory factor index IL-1β (Biovision, K4796-100) and the Xanthine Oxidase Fluorometric Assay Kit (Cayman Chemical, Ann Arbor, MI, USA) in the liver. The manufacturer’s instruction was followed and a VERSA Max microplate reader was used for measurement (Molecular Devices, San Jose, CA, USA).

Measuring Adenosine Metabolism in BALF

Check if the same lab product or an alternative is used in the 5 most similar protocols
Adenosine metabolism in BALF was assessed by measuring hypoxanthine generated after incubating 5 µL BALF supernatant with 200 µM adenosine in 50 µL Tris pH 7.5 at 37°C for one hour. Resulting hypoxanthine was assessed using a reaction mix containing Amplex red, horseradish peroxidase, and xanthine oxidase from the Xanthine Oxidase Fluorometric Assay Kit (Cayman Chemicals, Ann Arbor, MI). Signal was measured on a fluorometric plate reader with excitation 530 nM/absorbance 570 nM.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
