Polyclonal IgG Purification and Transfer

Check if the same lab product or an alternative is used in the 5 most similar protocols
Protein A/G is a recombinant protein expressed in Escherichia coli (50.5 kDa; 6 IgG binding sites) and is recommended for purifying polyclonal IgG from many species. Polyclonal mouse IgG from the serum of 7-9-week-old C57BL/6, non-lupus-prone MRL/+ mice and lupus-prone MRL/lpr mice or 6-7-month-old lupus-prone MRL/lpr mice was purified by Pierce Recombinant Protein A/G (Thermo Scientific Cat #21186). 100 μg IgG per mouse were i.v. injected into 7-9-week-old non-lupus-prone mice. PBS was used as the control.

Protein A/G Microarray Fabrication

Check if the same lab product or an alternative is used in the 5 most similar protocols
The microarray test sample was fabricated using the commercially available Pierce Recombinant Protein A/G (Thermo Fisher Scientific, 21186). First, the stock solution at 5 mg/ml was diluted using Milli-Q water to a final concentration of 500 μg/ml. This was then spotted (using a SCIENION sciFLEXARRAYER S3 spotter) onto a borosilicate glass slide (NEXTERION Slide E, SCHOTT), coated with a multipurpose epoxysilane layer that covalently binds most types of biomolecules including amino- and nonmodified DNA, RNA, and proteins. Spots of diameter 500 μm were made with 1000-μm center-to-center spacing, and the sample was left to dry overnight (24 hours) before measuring.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
