Immunostaining of p47phox in INS-1 cells

Cultured INS-1 cells were fixed in 4% paraformaldehyde. After washing and blocking, the cells were incubated with a rabbit anti-rat p47 phox monoclonal antibody overnight at 4 °C. After washing, the slides were incubated with FITCconjugated goat anti-rabbit immunoglobulins (DakoCytomation) for 45 min at 37 °C, washed, stained with PI (Sigma Chemical) and observed under a confocal microscopy (Leica TCS SP2 AOBS; Leica Microsystems, Cambridge, UK).
