In Vitro Toxicology Assay for Balsamin Purification

Assessing Caspase-8 Activity in MVEC Exposed to COVID-19 Plasmas

MVECs, 1.5 × 105/well, were cultured in 12 well 0.1 % gelatin-coated plates and incubated overnight at 37 °C in serum-free medium. The next day the cultures were overlain with control or patient plasmas (2 % v/v) in the presence or absence of varying concentrations of defibrotide or medium for ~5 h at 37 °C. Others had shown that very brief (1 h) exposure of primary human pulmonary MVEC to much higher concentrations (25 % v/v) of critical COVID-19 patient plasmas is cytotoxic [31] (link). Caspase 8 activity was assessed using a functional assay based on hydrolysis of Acetyl-Ile-Glu-Thr-Asp-p-nitraniline, performed according to the manufacturer's instructions (Caspase 8 Assay Kit, colorimetric; Sigma, St. Louis, MO), with measurement of optical density (OD) absorbance at 405 nm.
