Optimizing Cucumber Growth Under Cold Stress

Cucumber (Jinyan No. 4) seeds were sterilized in 2.5% NaClO and washed three times for 5 min in sterile distilled water, then soaked in distilled water for 6 h at 28°C. After the seeds were germinated on filter paper in Petri dishes, they were transferred to the growth chamber filled with vermiculite maintained at 28°C/18°C (day/night) with a 12 h photoperiod (photosynthetically active radiation = 400 μmol/m2/s). Cold stress condition maintained at 10°C/ 8°C (day/night), other conditions remain unchanged. Control, T1, T2, T3, T4, and T5 are the treatment of 0, 1, 10, 50, 100, and 1,000 μM sodium selenite (Sinopharm Chemical Reagent Co., Ltd), respectively. The concentration of exogenous application of sodium selenite was 1 μM under cold stress and normal conditions in the follow-up trial. The concentrations of melatonin (Tokyo Chemical Industry) and CPA (Tokyo Chemical Industry) were 1 μM each.

Melatonin (>98.0% purity) was supplied by Tokyo Chemical Industry (TCI, Tokyo, Japan). Iron (III) acetylacetonate (Fe[acac]3), polyvinyl alcohol (PVA, 87%–90% hydrolyzed, wt. 30,000–70,000) and PBS (pH 7.4) were purchased from Sigma-Aldrich (St Louis, MO, USA). Methylbenzene, oleylamine, and dichloromethane (DCM) were obtained from Sinopharm Group Co., Ltd. (Beijing, China). PLGA (50:50, wt. 90,000) was purchased from Jinan Daigang Biomaterial Co., Ltd. (Shangdong, China). High-glucose DMEM (H-DMEM), Roswell Park Memorial Institute 1640 medium (RPMI 1640), fetal bovine serum (FBS), penicillin–streptomycin (PS), trypsin-EDTA (0.25%) were purchased from Gibco (Life Technologies, Carlsbad, CA, USA). Cell Counting Kit-8 (CCK-8 assay) were obtained from Dojindo Molecular Technologies Inc. (Kumamoto, Japan). Rhodamine phalloidin and Coumarin-6 were supplied by Melone Pharmaceutical Co., Ltd. (Dalian, China). Deionized (DI) water was used throughout the study. All other chemicals and reagents were of analytical grade from commercial sources.

JNK-IN-8 ( JNK inhibitor) was obtained from EMB Chemical, Inc. (San Diego, CA). TMRM, Rhod-2 AM, HE and DCFH-DA stains were purchased from Molecular Probes, Eugene, OR. Hoechst 33258, SYTOX, N-acetylcysteine (NAC; ROS scavenger), tauroursodeoxycholic acid (TUDCA; ER stress inhibitor) and all other reagents were purchased from Sigma Aldrich (St. Louis, MO) unless otherwise specified. Melatonin and phenylarsine oxide are prepared in DMSO, therefore the amount of DMSO in each sample was 0.2%. However, while using the inhibitors, the maximum amount of DMSO utilized in the sample was also added as a vehicle in the control non-treated and other samples to equalize the effect.
