Histological Preparation of Rat Liver

Immediately after removal, rat liver portions were fixed for 1 day at room temperature in Carnoy’s fixative (ethanol:chloroform:acetic acid, 6:3:1), then preserved in 70% ethanol. Liver specimens were dehydrated with a graded series of alcohol preparations and embedded in paraffin. Specimens were cut in 7 μm sections using a rotary microtome and mounted on 3-aminopropyltriethoxysilane-coated glass slides. Each section was de-paraffinized in xylene, rehydrated in decreasing concentrations of alcohol in water, and stained with hematoxylin-eosin reagent (Sigma).
