Sections were triple-labeled with rabbit anti-NeuN (Millipore ABN78, 1/500), mouse anti-NMDAR2B (Abcam ab28373, 1/500), and goat anti-NMDAR2A (abcam ab118587, 1/500). Secondary antibodies were anti-rabbit-alexa-488, anti-goat-alexa-568, and anti-mouse-alexa-647 (Abcam ab150073, ab175474, and ab150107, respectively, 1/1000).

Quantitative Immunohistochemistry for GRIN2A/B

Check if the same lab product or an alternative is used in the 5 most similar protocols
The same animals from in situ hybridization were used for immunohistochemistry. Fixed sections were blocked using BLOXALL (catalog # SP-6000, Vector Laboratories), rinsed, and incubated in normal blocking serum. Sections were then incubated in 1:500 primary antibody (anti-GRIN2A, catalog # Ab118587; anti-GRIN2B, catalog # Ab65783, Abcam) overnight at 4°C. They were then incubated with appropriate secondary antibodies followed by avidin-biotin complex reagent (catalog # PK-6100, Vector Laboratories). Finally, they were washed and incubated in 3,3′-diaminobenzidine (DAB) peroxidase substrate (catalog # SK-4100, Vector Laboratories) and coverslipped. They were quantified the same way as in the in situ hybridization analysis.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
