Illustra ready to go pcr beads

Manufactured by GE Healthcare
Sourced in United States

Illustra Ready-To-Go PCR Beads are a pre-formulated reagent mix for polymerase chain reaction (PCR) amplification. Each bead contains all the necessary components for a single PCR reaction, including DNA polymerase, dNTPs, and reaction buffer. The beads are designed to simplify PCR setup and reduce the risk of contamination.

Genomic DNA Extraction and Amplification for Phylogenetic Analysis

Check if the same lab product or an alternative is used in the 5 most similar protocols
Genomic DNA was extracted from liver samples following the protocol used by Medeiros et al. [17 (link)]. The H1 and cytb fragments were amplified by PCR using the Illustra Ready-To-Go PCR Beads (GE Healthcare, Chicago, IL, USA), the primers MVZ59/MVZ50 [27 (link)] and 12SL13 [28 (link)]/16Sbr [25 ] in the case of the H1 fragments, and the primers MVZ15 [29 (link)] and H15149(H) [30 (link)] for the amplification of the cytb fragments. The resulting amplicons were purified using the Wizard SV Gel and PCR Clean-Up System (Promega, USA), and sequenced with the same primers used for PCR. In the case of the amplicon obtained with the primers 12SL13 and 16Sbr, the following primers were also employed for sequencing: H1KR [31 ], Hedges16SL2a [32 (link)], Hedges16SH10 [32 (link)], and 16Sar [25 ]. Sequencing reactions were prepared using BigDye Terminator (Applied Biosystems, USA), according to the manufacturer’s instructions. Nucleotide sequences were obtained by an ABI PRISM 3130XL Genetic Analyzer automated sequencer (Hitachi, Tokyo, Japan) at the DNA Sequencing Facility of the Chemistry Institute of University of São Paulo.

Genomic DNA Extraction from Kelengkeng Fruit Isolate

Check if the same lab product or an alternative is used in the 5 most similar protocols
Leuconostoc mesenteroides strain K7 isolated from Kelengkeng fruit was used as the genomic DNA source. All of the analytical and molecular biology grade chemicals used in this study, namely, nutrient agar and nutrient broth (Sigma Aldrich), DNA ladder (Vivantis), DNA loading dye, Illustra™ Ready-To-Go PCR beads (GE Healthcare), nuclease-free water (Gibco), PureLink™ Quick Gel Extraction Kit (Life Technologies), and BigDye XTerminator Kit (Applied Biosystems), were commercially available.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
