Five micron sections of tumor biopsies were subjected to immunohistochemistry using Ventana reagents and a Ventana autostainer in a CLIA histology laboratory. Immunohistochemical Analysis of IPF Lung

Serial sections of paraffin-embedded parenchymal lung tissue from IPF patients were immunolabelled for p21 using a rabbit polyclonal antibody (Cell Signaling Technology, Danvers, MA, USA, #2947, diluted 1 in 50) and detected using the Dako EnVision anti-rabbit kit (Dako Corp., Carpinteria, CA, USA) and 3,3′-diaminobenzidine (DAB, Sigma-Aldrich, St Louis, MO, USA); or stained with Masson’s Trichrome to identify collagen. Masson’s trichrome staining was performed at The Hunter Medical Research Institute’s (HMRI’s) Pathology Service laboratory and 40× magnification images digitalised using the Aperio AT2 digital scanner (Leica, Wetzlar, Hesse, Germany).

Histological Evaluation of Distal Colon

Distal Colons were fixed in 10% formalin and stained with routine H&E. Histology was scored in a blinded fashion as described previously.29 (link) Briefly, histology scores include a combination of epithelium morphology change (score 0–4; same below), inflammatory cell infiltration/inflammation and mucosal ulceration. Score 1–4 indicated that the damage was observed in <25%; 25-50%; 50-75% and 75% area, respectively, while score 0 indicated normal condition. A total number of four H&E sections per animal were assessed for each sample and the average scores were used. Finally, the sum scores were determined by the combined numbers of all three parameters, representing the overall histopathology status of each sample. Representative images were acquired with a Leica AT2 digital scanner. Histology images with a score close to group average were displayed as examples.
