All other reagents used were analytically pure.

Characterization of Fungicide Compounds

Analytical grade compounds, including SDHIs (bixafen, boscalid, fluopyram, fluxapyroxad and penthiopyrad), fentin chloride, chlorothalonil and tolnaftate, were all purchased from Sigma‐Aldrich (UK). Isofetamid (N‐[1,1‐dimethyl‐2‐(4‐isopropoxy‐o‐tolyl)‐2‐oxoethyl]‐3‐methylthiophene‐2‐carboxamide) was synthesized and supplied with >95% purity by Nihon Nohyaku Co. Ltd. (Japan). Chemicals were dissolved in pure dimethyl sulfoxide (DMSO) at 10 mg mL−1 and stored at −20 °C before further use.

Antifungal and Immune Elicitor Compounds

Saponins aescin (E1378), hederagenin (H3916), and soyasaponin I (S9951), and fungicides metconazole, fluconazole, boscalid, and fluopyram (all purchased from Sigma–Aldrich, St. Louis, MO, USA) were dissolved in 99.8% ethanol (EtOH) as 10 mg·ml−1 stock solution. Tebuconazole, in the form of the commercially formulated product Horizon 250 EW (Bayer CropScience, Germany), was also prepared as 10 mg·ml−1 stock solution in EtOH. Ergosterol (Sigma, St. Louis, MO, USA) was prepared as 5 mM stock solution in EtOH and used at the final concentration of 25 µg·ml−1. Benzothiadiazole (BTH) was used in the form of the commercially formulated product Bion 50 WG (Syngenta, Switzerland) and prepared directly into the working solutions. The commercial peptide flg22 (EZBiolab) was diluted in Milli-Q water and used at the final concentration of 1 µM. All stock solutions were stored at −20°C.

Assessing Nematode Fertility under Pesticide Exposure

Synchronized L4 hermaphrodite worms (N2, RP2698, and RP2674) were individually placed on NGM plates seeded with E. coli OP50 and exposed to various concentrations of cyclobutrifluram or fluopyram (Sigma-Aldrich, Burlington, MA, USA). The worms were transferred to new plates every 24 h until egg laying stopped. The total number of eggs, adult hermaphrodites, and males were scored. The average brood size, embryonic viability, and incidence of males were determined. Statistical significance was assessed using a two-tailed Student’s t-test with Welch’s correction, p-value ≤ 0.05.

Comprehensive Analysis of Fungicide Residues

Seventeen agricultural fungicides (test range 32–0.03 mg/L), five azole fungicide metabolites (test range 16–0.016 mg/L) and four medical azoles were included (test range 32–0.03 mg/L). The agricultural fungicides (bixafen, boscalid, fluxapyroxad, fluopyram, folpet, azoxystrobin, prothioconazole, paclobutrazole, epoxiconazole, propiconazole, tebuconazole, difenoconazole, metconazole, prothiozonacole-desthio, prochloraz, imazalil were purchased from Sigma-Aldrich, Søborg, Denmark, whereas mefentrifluconazol was purchased from LGC Standards (Teddington, Middlesex, United Kingdom www.lgcstandards.com, accessed on 26 January 2021). The metabolites included: 1,2,3-triazole (Sigma-Aldrich, Søborg, Denmark), 1,2,4-triazole and triazole sulfonamide (Toronto Research Chemicals, Toronto, Canada), and triazole alanine and triazole acetate (HPC Standards GmbH, Borsdorf, Germany). The medical antifungal agents used were itraconazole, voriconazole, isavuconazole and posaconazole (all from Sigma-Aldrich, Søborg, Denmark, except isavuconazole, which derived from Basilea).

Pesticide Standards Quantification Protocol

The pesticide standards that included acetamiprid, boscalid, cyprodinil, dimethoate, dithianon, fenhexamid, fenoxycarb, fenpyrazamine, fludioxonil, fluopyram, iprodione, pyraclostrobin, pirimicarb, tebuconazole, thiacloprid, and trifloxystrobin with a purity in the range of 99.4–100 % were purchased from Sigma Aldrich (Germany). The chemical structures selected compounds are shown in Figures S13-S14. Stock solutions were prepared in acetonitrile and stored at −20 °C. Deionized water for the preparation of the mobile phase was produced by Watek system (Germany). Ammonium formate for mass spectrometry was obtained from Fluka (Germany). Formic acid used in the mobile phase and acetonitrile in HPLC grade was obtained from Sigma-Aldrich (Germany). Methanol (HPLC grade) was from Merck (Germany).
