ChIP Assay for ZEB1 and TFAP2C

Cells were harvested for chromatin immunoprecipitation (ChIP) by using an EZ-ChIP kit based on the manufacturer’s instructions (Millipore, Burlington, MA, USA). ZEB1 and TFAP2C were obtained from Abcam. The ChIP primer sequences are listed in Table S1. Immunoprecipitated DNA was quantified using qPCR. Purified DNA was quantified using real-time qPCR.

Western Blot Analysis of Cell Signaling Proteins

Cold lysis buffer containing 50 mmol/l Tris–HCl (pH 7.4) was used to lyse the obtained cells. The DC protein assay kit (Bio-Rad Laboratories) was used to test the protein content, and a western blot was conducted according to the scheme of Hnasko's protocol. [23 ] Antibodies TFAP2C, TP53, P21, EGFR, Cylin D1, Bcl2, Bax, cCaspase-3 and β-actin were all bought from Abcam (Cambridge, UK). The enhanced chemiluminiscence solution (Life Sciences; Thermo Fisher Scientific) was used to visualize proteins.

IHC Analysis of Subcutaneous Tumor Samples

The collected subcutaneous tumors were fixed with 4% formalin, embedded in paraffin and then sectioned into 4-μm-slices. The standard protocol for IHC of the TMAs and subcutaneous tumors using a streptavidin-peroxidase (SP) Kit (Zhongshan biotech, Beijing, China), as described previously [25 (link)]. The slides were incubated with antibodies specific for TFAP2C (Abcam, cambridge, MA, USA), TRIM29 (Proteintech, Chicago, IL, USA), PHLPP1 (Proteintech, Chicago, IL, USA), p-AKT (Cell Signaling Technology, Danvers, MA, USA) and Ki-67 (Cell Signaling Technology, Danvers, MA, USA). The images of IHC staining were obtained by an Olympus microscope (Tokyo, Japan).
