Quantification of Herbal Powder Formula

A reference standard of spironolactone was purchased from Research Biochemicals International. Canrenone (purity ≥97%) and the internal standard n-propylbenzene were obtained from Sigma-Aldrich Inc. All chemicals were of analytical grade, including formic acid (98–100%) and ethanol, which were purchased from E. Merck. The pharmaceutical herbal powder formula Yin-Chen-Hao-Tang was purchased from Kaiser Pharmaceutical Co., Ltd. and was quantified by a Shimadzu UHPLC system coupled to an electrospray ionization (ESI) source equipped with an LCMS-8030 triple quadrupole mass spectrometer to guarantee the consistent quality of the herbal formula powder [33 ].

Steroid Receptor Activation Assay

Cell culture materials are from Life Technologies (Cergy-Pontoise, France). Luciferin (sodium salt) was purchased from Promega (Charbonnières, France). Chemical substances used in this study are presented in Table 1. Methyltrienolone (R1881), dexamethasone (DEX), mifepristone (RU486), aldosterone (ALDO), pregnenolone (P5), progesterone (P4), dydrogesterone (DYD), norethindrone (NET), tibolone (TIB), spironolactone (SPI), canrenone (CAN), fluticasone propionate (FT), deacetyl cortivazol/bimedrazole (DAC), cortivazol (CVZ), methylprednisolone (MPS), cortisol (CORT), dihydrotestosterone (DHT), drospirenone (DRO) and17α-hydroxyprogesterone (17-OHP) were obtained from Sigma-Aldrich (Saint-Quentin Fallavier, France). Promegestone (R5020), forbimenol, RU24782 and RU24858 are synthetic steroids non-commercially available and kindly gifts from Sanofi, Vertolaye, France.
Stock solutions of chemicals were prepared in dimethyl sulfoxide (DMSO) and stored at -20°C. Fresh solution of test chemicals in test medium were prepared before each experiment. The final DMSO concentrations during treatment did not exceed 0.1% (v/v) of the test medium.

Quantitative LC-MS Analysis of Spironolactone

Spironolactone and canrenone were purchased from Sigma Aldrich (St. Louis, MO, USA), 7-alpha-methylthioSpironolactone from PharmaSynth (Tartu, Estonia), and Spironolactone-D3 (used as an internal standard, IS) from TLC Pharmaceutical Standards (Aurora, Ontario, Canada). LC-MS-grade methanol, tetrahydrofuran (THF), NH4F, and 1,1,1,3,3,3-hexafluoro-2-propanol (HFIP) were obtained from Sigma Aldrich (St. Louis, MO, USA). Water was purified (18.2 MΩ·cm at 25 °C and a TOC value in the range 2–5 ppb) in house using a Millipore Advantage A10 system from Millipore (Bedford, USA).
