Bronchial Epithelial Cell Culture and Treatment

BEAS-2B bronchial epithelial cells (ATCC, Rockville, MD) were maintained with complete BEGM media (Lonza, Basel, Switzerland) supplemented with 10% FBS and plated on dishes coated with collagen-fibronectin-BSA mixture. Calu-3 cells (ATCC) were maintained in EMEM media supplemented with 10% FBS plated on 3 μM polyester membrane transwell supports (Corning, Corning, NY). After plating cells, epithelial tight junctions were allowed to mature over 7–10 days until the TER was over 1000 Ω x cm2. Cells were treated with 3-oxo-C12-HSL (Sigma-Aldrich, St. Louis, MO), trans-resveratrol (Cayman Chemical, Ann Arbor, MI), or metformin (Cayman Chemical), or co-cultured with the P. aeruginosa strain, PAO1, (MOI of 1 or 30), for 6 or 16 hours. Primary NhBE cells were grown in conditional reprogrammed cell (CRC) culture medium as described in Liu et al.42 (link), on human type IV placental collagen-coated 12-mm Costar Transwell supports (Corning) with slight modifications, by the CF@LANTA RDP Experimental Models Core at Emory University as described previously43 (link).

Trans-resveratrol Dosing and Administration in Rats

Trans-resveratrol (Cayman Chemical Company, Ann Arbor, MI) was dissolved in a solution consisting of 10% ethanol (Ultra Pure, Darien, CT), 15% Cremophor (Fisher Scientific, Waltham, MA), and 75% saline (Abbott Laboratories, Chicago, IL) to a concentration of 10 or 30 mg/ml. These doses were chosen as 10 and 30 mg/kg RSV treatment selectively produces peripheral anti-inflammatory effects (Hong et al., 2008 (link); Singleton et al., 2010 (link)), yet only the higher dose of 30 mg/kg accesses the brain as identified by high performance liquid chromatography studies (Wang et al., 2002 (link)). Therefore, vehicle (0), 10, or 30 mg/kg RSV (ip) was administered to rats beginning 7 days before control/defeat exposure and ending on the last day of defeat. All tissue was collected 5 days after the final treatment with RSV (see Figure 1 for a brief study timeline). Since the half-life of RSV following repeated dosing is reported to be 2.5 hours, RSV is not likely to persist in plasma or brain tissue at time of collection (Almeida et al., 2009 (link)).

Antioxidant Characterization of Brazilian Wine

Wine alcohol 65% v/v was obtained from winery Ouro Verde-Miolo Wine Group (Casa Nova, BA, Brazil). Folin-Ciocalteu reagent, ethanol, potassium persulfate, phosphoric acid, and potassium phosphate monobasic were purchased from Merck (Darmstadt, Germany). External standards of gallic acid, syringic acid, benzoic acid, p-coumaric acid, o-coumaric acid, cinnamic acid, procyanidin B1, catechin, procyanidin B2, and caffeic acid came from Sigma-Aldrich (St. Louis, MO, USA). Epigallocatechin gallate, epicatechin, epicatechin gallate, procyanidin A2, quercetin 3-glucoside, rutin, kaempferol 3-glucoside, myricetin, and isorhamnetin 3-glucoside, quercetin-3-O-glucopyranoside came from Extrasynthese (Genay, France). The trans-Resveratrol was obtained from Cayman Chemical Company (Michigan, EUA, Ann Arbor, MI, USA).

Investigating Lifespan Effects of Bioactive Compounds on Flies

We greased fly medium by paste of hydrolyzed yeast containing one of the substances. Control untreated animals were fed by yeast past without substances. To make the hydrolyz at yeast were boiled in water bath for 30 minutes. To prepare the 100 ml of paste 50 g of dry yeast per 60 mL of water were used.

Polyphenol Standards and Isotope-Labeled Compounds

(Chuo-ku, Tokyo, Japan). A 0.1 N standard solution of hydrochloric acid was purchased from Dae-Jung Chemicals & Metals Co., Ltd. (Shiheung-city, Gyeonggi-do, Korea), and deionized water with a resistivity of 18.2 MΩ was obtained from a PURELAB Option-Q System (Lane End, High Wycombe HP14 3BY, UK).

The ultrapure water was obtained using a Marte Científica purification system (São Paulo, SP, Brazil). The chromogen 3,3′diaminobenzidine tetrachloride was purchased from Vector Laboratories (Burlingame, CA, USA). All antibodies used in this study were purchased from Abcam (Cambridge, MA, UK). All chemical reagents used in the experiments were of analytical grade.
