Protein Detection via Western Blotting

Exogenously expressed proteins ANP32B and PB2, and the cellular control protein Vinculin were probed with primary antibodies rabbit anti-ANP32B (Abcam, ab200836; 1/2000), rabbit anti-influenza A virus PB2 (Genetex, GTX125925; 1/2000) and rabbit anti-Vinculin (Abcam, ab129002; 1/2000), respectively. The backsides of the primary antibodies were then detected with goat anti-rabbit secondary antibody conjugated to horseradish peroxidase (Genetex, GTX213110-01; 1/10,000). Detection was carried out using either Cytiva ECL Start Western Blotting Detection reagent (Amersham) or Immobilon Western chemiluminescent HRP substrate (Millipore).

Western Blot Analysis of Actin, FLAG, PHAP1, and PHAPI2

For Western blot analysis, cells were lysed in NP-40 lysis buffer containing 1× cOmplete Protease Inhibitor Cocktail (Roche) and 1× Phenylmethylsulfonyl fluoride (Sigma-Aldrich) and cleared from the insoluble fraction by centrifugation at 17,000 × g for 5 min at 4°C. Samples were analyzed by SDS-PAGE and transferred onto nitrocellulose membranes. Membranes were probed with mouse monoclonal anti-Actin (Thermo Scientific, MS-1295), mouse monoclonal anti-FLAG M2 (Sigma-Aldrich, F1804), rabbit polyclonal anti-PHAP1 (Abcam, ab51013), and rabbit monoclonal anti-PHAPI2 (Abcam, ab200836) antibodies. Primary antibodies were detected using HRP-conjugated secondary anti-mouse (GE Healthcare, NA931V) and anti-rabbit (GE Healthcare, NA934V) antibodies and visualized using Immobilon Western Chemiluminescent HRP substrate kit (Millipore) according to the manufacturer’s instructions.

Immunoblotting of ANP32 Proteins

A glycogen periodic acid-Schiff (PAS) staining kit was purchased from Beijing Solarbio Science & Technology Co., Ltd (Beijing, China). LPS was obtained from sigma (Code No L4391-1MG). TAK-242 (resatorvid) was purchased from MedChemExpress.
