All chemicals used in the syntheses unless otherwise specified were purchased from Sigma-Aldrich Co. LLC. l-lysine and DiS-C3-(5) were purchased from Combi-Blocks, Inc. N-(3-dimethylaminopropyl)-N′-ethylcarbodiimide hydrochloride (EDC.HCl) and 1-hydroxybenzotriazole (HOBt) were purchased from GL Biochem Ltd. Cyclophosphamide was purchased from MedChemExpress LLC. PI and FM 4–64FX were purchased from Thermo Fisher Scientific. 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) was purchased from Alfa Aesar. FITC was purchased from Biotium, Inc. Technical grade solvents were purchased from SG Labware Pte Ltd. (Singapore) and directly used for column purification without any distillation. Pullulan standards were purchased from Polymer Standards Service.

Oligonucleotide Primers were custom synthesized from Sigma-Aldrich, USA. PCR reagents, Taq Polymerase (Platinum HiFi) was obtained from Invitrogen, USA, Plasmid Miniprep kits, Gel extraction buffers, PCR purification kits, Ni-NTA beads, were obtained from Qaigen,USA. pET23b plasmid, T7 promoter /terminator primer was supplied by Novagen Inc., USA. All other chemicals and solvents used in the study were obtained from Sigma-Aldrich, USA.

Coupling reagents such as N,N,N′,N′-tetramethyl-O-(1H-benzo-triazol-1-yl)uronium hexafluorophosphate (HBTU) and 1-hydroxybenzotriazole (HOBt), as well as the Rink amide resin used, all were commercially obtained by GL Biochem Ltd. (Shanghai, China). N,N-diisopropylethylamine (DIEA), piperidine, and tri-fluoroacetic acid (TFA) were purchased from Iris-Biotech GMBH. Peptide synthesis solvents and reagents, such as N,N-dimethylformamide (DMF), dichloromethane (DCM), diethyl ether (Et2O), water and acetonitrile (MeCN) for HPLC, were reagent grade acquired from commercial sources (Sigma-Aldrich and VWR) and used without further purification.

AIBN was purified by recrystallization in ethanol according to the literature [23 (link)]. 1-(3-Dimethy laminopropyl)-3-ethylcarbodiimide HCl (EDC·HCL) and 1-hydroxybenzotriazole (HOBT) were purchased from GL Biochem (Shanghai, China). Dulbecco’s Modified Eagle’s Medium (DMEM) and 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) was bought from Sigma-Aldrich (MO, USA).

The animal-related experiments were performed in full compliance with institutional guidelines and approved by the Animal Use and Care Administrative Advisory Committee at the University of Pittsburgh. Informed consent was obtained for experimentation with patient-derived xenograft (PDX) model. And the PDX-related experiments were carried out in accordance with The Code of Ethics of the World Medical Association.
