For self-administration experiments in mice and rats, nicotine bitartrate dihydrate (CAS# 6019–06-3, MP Biomedicals, Santa Ana, CA) was dissolved in 0.9% sterile saline. All doses of nicotine refer to the free-base form. Atenolol (CAS# 29122–68-7, Sigma-Aldrich, St. Louis, MO) was dissolved in 2mM HCl in 0.9% sterile saline. The GLP-1 receptor agonist Ex-4 (Tocris, Ellisville, MO) was dissolved in 0.9% saline solution for in vivo use and 0.1M PBS for ex vivo use. Clozapine-N-oxide (CNO, Enzo Life Sciences, Farmingdale, NY), insulin (CAS# 11070–73-8, Sigma-Aldrich, St. Louis, MO) and glucagon (CAS# 16941–32-5, Sigma-Aldrich, St. Louis, MO) were diluted in 0.9% saline for intraperitoneal injection. D-(+)-Glucose (CAS# 50–99-7, Sigma-Aldrich, St. Louis, MO) was dissolved in dH2O for oral gavage. cAMPS-RP, 8-Br-cAMP, and Rolipram (Tocris, Ellisville, MO) were all dissolved in 0.9% saline. The pH of all solutions was adjusted to ~7.4. The pH of all solutions was adjusted to ~7.4.

Menthol and Nicotine Pharmacological Effects

L-(−)-Menthol (Fisher Scientific, Atlanta, Georgia, USA) was dissolved in 50% dimethyl sulfoxide (DMSO; Sigma; Atlanta, Georgia, USA) and distilled water (v/v). Menthol and vehicle (50% DMSO: H2O, v/v) solutions were freshly prepared weekly and injected intraperitoneally (IP) at 1 ml/kg with a 5-min IPI. All menthol and vehicle solutions were heated to 50°C before each session to ensure menthol solubility. All nicotine and menthol training doses were selected based on previous literature (Biswas et al., 2016 (link); Huynh et al., 2019 (link)). (−)-Nicotine bitartrate dihydrate (MP Biomedicals; Solon, Ohio, USA) was dissolved in 0.9% saline, titrated with NaOH to a pH of 7.0 ± 0.2, and injected subcutaneously (SC) at 1 ml/kg with a 5-min IPI. All nicotine doses are expressed as the free base form and all menthol doses are expressed as the salt form.

Nicotine bitartrate dihydrate (MP Biomedicals LLC, catalog no. 0521149910) was dissolved in saline and the pH adjusted to 7.6 with NaOH. Nicotine solutions were prepared daily in glass vials wrapped in aluminum foil to protect from light and sterilized by being passed through a 0.25-µm syringe filter (Fisher Scientific, catalog no. 09-719C). Nicotine doses are reported as free base. Recombinant HHIP protein was purchased from R&D systems (catalog no. 1568-HP). Murine Shh protein (catalog no. 315-22) and human Shh protein (catalog no. 100-45) were purchased from Preprotech and used at a final concentration of 3 µg⋅mL−1.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
