Microarray Analysis of lncRNA Expression

Check if the same lab product or an alternative is used in the 5 most similar protocols
Microarray analysis was conducted as a previous report [18 (link)]. In brief, total RNA from 5 pairs of CC and paracancerous tissues was extracted. Next, cDNA was synthesized using 0.5 μg total RNA via a GeneChip 3’In-vitro Transcription Express Kit (Thermo Fisher Scientific Inc., Waltham, MA, USA, 902789). Then the cDNA was fragmented and hybridized with human LncRNA Array V3.0 (Arraystar Inc., USA, AS-LNC-H-V4.0). After that, the chips were washed and scanned using a GeneChip™ Scanner 3000 7G system (Thermo Fisher, 000213).

Profiling lncRNA Expression in CRC Serum EVs

Check if the same lab product or an alternative is used in the 5 most similar protocols
Microarray analysis was performed following the steps described previously in the literature [16 (link)]. In brief, total RNA was extracted from the serum EVs from 6 CRC patients and 6 healthy subjects. Subsequently, total RNA from the EVs was extracted and 0.5 μg RNA was used for cDNA synthesis using a GeneChip 3′. In vitro transcription express kit (902789, Thermo Fisher Scientific, Waltham, MA, USA). Subsequently, the cDNA was segmented and hybridized with a human lncRNA expression array V3.0 (AS-LNC-H-V4.0, Arraystar Inc. Rockville, MD, USA). After hybridization, the microarray was washed and scanned with a GeneChip Scanner 3000 7G System (000213, Thermo Fisher Scientific).

Differential lncRNA Expression Analysis

Check if the same lab product or an alternative is used in the 5 most similar protocols
Microarray analysis was performed to screen out the differentially expressed lncRNAs between tumor and normal tissues. In brief, 0.5 µg total RNA from either tumor tissues or normal tissues was reversely transcribed to cDNA using a GeneChip 3'In vitro Transcription Express Kit (Thermo Fisher Scientific, Waltham, MA, USA). Then the cDNA was hybridized with Human LncRNA Expression Array V4.0 (Arraystar, Rockville, MD, USA), and then the chip was washed and scanned on a GeneChipTM Scanner3000 7G System (Thermo Fisher).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
