The Poly6 (GRLVFQ) peptide from the HBV polymerase region overlapping with the preS1 deletion region, mainly discovered from occult HBV infection or severe liver disease progressive patients, was synthesized by the (9-fluorenylmethoxycarbonyl Fmoc)-based solid-phase method and characterized by Peptron Inc. (Cheongju, Korea) (Figure 1A and Figure S1).
Remdesivir (GS-5734, RDV), hydroxychloroquine sulfate (S4430, HCQ), and ruxolitinib (INCB018424) were purchased from Selleck Chemicals (Selleck, Houston, TX, USA). Unless otherwise indicated, all other chemicals were purchased from Sigma-Aldrich (St. Louis, MO, USA).

Cells were harvested in lysis buffer (50 mM Tris-HCl (pH 7.4), 150 mM NaCl, 1% NP-40, 0.25% deoxycholate, and a protease inhibitor cocktail (Roche, Basel, Switzerland)). The lysates were then subjected to 12% SDS-PAGE and Western blotting according to our standard procedures [18 (link),44 (link)]. The antibodies used in this study included anti-tubulin (1:3000), anti-His (1:10,000), anti-Flag (1:5000), and anti-Dicer (1:2000), which were purchased from Proteintech, Rosemont, IL, USA. The JAK1/JAK2 inhibitor Ruxolitinib (INCB018424) was purchased from Selleck, Houston, TX, USA (CAS No. 941678-49-5).

Versican monoclonal antibody (12C5) recognizing the G1 hyaluronan-binding domain of human versican was purchased from Developmental Studies Hybridoma Bank (Iowa University), and VCAN polyclonal antibody (ab19345) recognizing the ADAMTS-1/4 cleavage site (aa 436–441) was purchased from Abcam (San Francisco). The flow cytometry antibodies to pSTAT3 (Tyr(P)-705) and to pSTAT5 (Tyr(P)-694), conjugated to PE and to Alexa Fluor 647, respectively, were purchased from BD Biosciences. The Src family kinase inhibitor PP2 was purchased from EMD Biosciences (San Diego, CA). The Src family kinase inhibitor dasatinib and the JAK inhibitor ruxolitinib (INCB018424) were purchased from Selleck Chemicals (Houston, TX). STAT3/5 inhibitor C188-9 was obtained from Dr. Michelle Redell (Baylor College of Medicine), and the STAT3/5 inhibitor 5,15-DPP was purchased from Abcam (San Francisco, CA). Puromycin dihydrochloride was purchased from Mediatech. Doxycycline was purchased from Clontech (Mountain View, CA).
