Comprehensive Reference Standards for Drug Analysis

The reference standards were obtained from BD Biosciences (Franklin Lakes, NJ, United States), Cayman Chemicals (Ann Arbor, MI, United States), Cerilliant (Round Rock, TX, United States), Chiron AS (Trondheim, Norway), LGC Standards (Wesel, Germany), Lipomed AG (Arlesheim, Switzerland), Sigma-Aldrich (Schnelldorf, Germany), Toronto Research Chemicals (North York, ON, Canada), USP (Rockville, MD, United States), Wyeth Pharmaceuticals LLC (Collegeville, PA, United States), and standards donated by the Slovenian National Forensic Laboratory. The internal standards (ISs) amphetamine-d5, diazepam-d5, methadone-d3, mianserin-d3, and morphine-d6 were purchased from Cerilliant (Round Rock, TX, United States). Acetonitrile, ammonium acetate, ammonium formate, formic acid, and methanol were of LC-MS grade and obtained from Fisher Scientific (Loughborough, United Kingdom).

Quantification of Morphine Sulfate in Plasma

Morphine sulfate concentrations were quantified in plasma, following solid-phase extraction, using a validated high-performance liquid chromatography (HPLC) method with tandem mass spectrometry (MS/MS) detection (AB Sciex API 5000 quadrupole mass spectrometer) in positive ion mode. A reversed-phase C18 column (50 × 2.10 mm, with 3.5 µm porosity) was used with two mobile-phase gradients (A: 20 mM NH4HCO3, pH 10.0; and B: ACN:MeOH 75:25 v/v) and a constant flow rate of 0.5 mL/min. Retention times for morphine (purity 99.6%) and morphine-D6 (purity 98.9%), used as internal standards (Cerilliant Corp., Round Rock, TX, USA), were 0.92 min and 0.88 min, respectively. Peak area ratios were used for quantification, and linearity of the calibration curve was determined using a 1/×2 weighted linear least squares (LS) regression analysis. The lower limit of quantification (LLOQ) of 50.0 pg/mL had a between-run accuracy of 91.4% and precision of 6.7%. All calibration and quality control samples were within the acceptance criteria: CV% < 15% and nominal range values within 85–115%.

Quantification of Opioids and Sedatives

Standard substances and their respective stable isotope labeled internal standards (IS): M3G, M6G, morphine, clonidine, MiOH, midazolam, M3G-D3, M6G-D3, morphine-D6 and MiOH-D4 were obtained from Cerilliant (Texas, USA). Clonidine-D4 and midazolam-D6 were obtained from the Toronto Research Chemicals Inc. (Toronto, Canada). Other reagents used: LC-MS Ultra chromasolv grade methanol (MeOH) from Sigma Aldrich (Missouri, USA), LC-MS grade formic acid from Sigma Aldrich (Missouri, USA), LC-MS grade ammonium hydroxide solution from Sigma Aldrich (Missouri, USA), LC-MS grade 1,1,1,3,3,3-hexafluoro-2-propanol (HFIP) from Sigma Aldrich (Missouri, USA). Water was purified (18.2 MΩ•cm at 25 °C and and a total organic carbon (TOC) value 2 -3 ppb) in-house using a Millipore Advantage A10 system from Millipore (Bedford, USA). Plasma and whole blood were purchased from Blood Bank of Tartu University Hospital.

Quantification of Illicit Drugs in Cough Syrups

Carbinoxamine, chlorpheniramine, and chlorpheniramine-d6 were purchased from TRC (North York, Canada). Individual solutions of 6-AM, 7-aminonitrazepam, 7-aminonimetazepam, amphetamine, benzoylecgonine, C6G, codeine, codeine-d6, ketamine, methamphetamine, mephedrone, 4-methylephedrine, 3,4-methylenedioxymethamphetamine hydrochloride, M3G, M6G, morphine, morphine-d6, and norketamine (each with a concentration of 1 mg mL−1) were purchased from Cerilliant (Round Rock, TX, USA). Three brands of cold syrups (Consrine, ssBuron, and Pabron) manufactured in Taiwan were purchased from local pharmacies. Consrine contains codeine phosphate (0.48 mg mL−1), chlorpheniramine (0.25 mg mL−1), dl-methyl ephedrine HCl (1.6 mg mL−1), and glycyrrhizic acid (0.25 mg mL−1), ssBuron contains codeine phosphate (0.5 mg mL−1), guaifenesin (3.33 mg mL−1), chlorpheniramine maleate (0.14 mg mL−1), and caffeine (1.33 mg mL−1), Pabron contains codeine phosphate (0.48 mg mL−1), dl-methyl ephedrine HCl (2.5 mg mL−1), and carbinoxamine maleate (0.4 mg mL−1). Formic acid and methanol (high-performance LC grade) (≥ 98%) were purchased from J.T. Baker (Pittsburgh, PA, USA). Ultrapure water (18.2 mΩ·cm) obtained from a Milli-Q ultrapure water system (Millipore, Billerica, MA, USA) was used in this study.
