Modeling BRAF-driven Cancers in Mice

All mice strains were obtained from the Jackson laboratory, which include BRAFCA/+ (#017837, a Cre-driven BRAF(V600E)-knockin strain) [58 (link)], Trp53flox/flox (#008462), pTENflox/flox (#006440), p27flox/flox (#027328), Mx1-cre (#003556), and cd19cre/+ (#006785) mice strains. These strains were bred to generate BRAFCA/+Mx1-cre, BRAFCA/+ cd19cre/+, BRAFCA/+Trp53flox/flox cd19cre/+, BRAFCA/+pTENflox/flox cd19cre/+, BRAFCA/+p27flox/flox cd19cre/+ mice strains. All mice strains were maintained in the animal facility at the National Cancer Center Singapore (NCCS). To track the disease progression in mice with different genetic modifications, 20 mice per strain were used and monitored up to 40 weeks. To characterize the pathological features of disease in mice, animals at the terminal stage of diseases were euthanized and sacrificed for harvesting of blood, spleen, liver, lung, skin and bone marrow according to standard protocols. All operations were approved by the institutional Animal Care and Use Ethics Committee of NCCS.

Genetically Engineered Mouse Models for GI Tumors

All mouse studies are conducted in accordance with MSKCC IACUC under protocol 11-12-029. The BrafCA, Etv1CreERT2, Trp53flox and Rosa26CAGGS-LSL-EYFP mice were obtained from The Jackson Laboratory. To trace the Etv1-expressing cellular compartments in the GI tract, Etv1CreERT2/+; Rosa26CAGGS-LSL-EYFP/+ mice were generated through standard mouse breeding with Etv1CreERT2/CreERT2 and Rosa26CAGGS-LSL-EYFP/CAGGS-LSL-EYFP mice. Etv1CreERT2; BrafCA/CA and Etv1CreERT2; BrafCA/+ mice were generated through breeding of Etv1CreERT2 and BrafCA/CA mice. To cross into the Trp53 conditional background, we crossed Etv1CreERT2;BrafCA/CA with Trp53flox mice to generate Etv1CreERT2;BrafCA/+;Trp53flox/flox and Etv1CreERT2;BrafCA/CA;Trp53flox/flox compound genetically engineered mice.
