Pepsin (P7125) is a proteolytic enzyme derived from porcine gastric mucosa. It has a core function of hydrolyzing peptide bonds in proteins, particularly those involving the carboxyl group of aromatic amino acids such as phenylalanine, tryptophan, and tyrosine.

This product was plasticized with polyethylene glycol 400, PEG 400, obtained from Merck-Schuchardt (Germany). The model drug, diclofenac sodium was obtained from Aarti drugs Ltd. (India) and was used as received. Diclofenac sodium was chosen on the basis that an extended release formulation of this, some are already available in market, can prove useful in management of signs and symptoms of patients with osteoarthritis and rheumatoid arthritis. Pepsin (p7125) and pancreatin (p8096) enzymes were purchased from Sigma-Aldrich (USA). Other materials used for the preparation of the dissolution media were reagents of standard grades and were supplied by AZ chem and Alpha Chemika (India).
