Sitosterol and Sitostanol Cell Exposure

Stock solutions of sitosterol and sitostanol (Sigma) were prepared in ethanol and provided to the different in vitro cell models. Final sitosterol and sitostanol concentrations used were 12 µM and 1.2 µM, respectively, and compared to ethanol as carrier control. All conditions were set at identical ethanol concentration of 0.25%.

Flaxseed Extraction and Characterization

Flaxseed was obtained from the Gansu Academy of Agricultural Sciences, China. The raw material was cleaned to remove dust, stone, other seeds, etc. The cleaned seed was dried at 50℃ until reducing to the moisture of approximately 6%. The dried sample was ground in a blender and then pass through a 30-mesh sieve. The ground seed was sealed and stored in the dark at 4℃ until use.
n-Butane solvent (99.5%) was obtained from Longyu Chemical Co., Ltd. (Puyang, China) . Fatty acids and phytosterols standards ( Campesterol, Stigmasterol, β-Sitosterol, Sitostanol and 5α-cholestanol) were purchased from Sigma-Aldrich Chemical Co., Ltd. (Shanghai, China) . All other chemicals used in study were at least of analytical grade.

Quantification of Oryzanol and Sterols

All solvents and chemicals were of analytical grade or HPLC/GC grade and purchased from RCI Labscan (Bangkok, Thailand). The total γ-oryzanol (98.5%) standard was purchased from Tsuno Rice Fine Chemical Co., Ltd. (Wakayama, Japan). PC standards, including docosanol (C22), tetracosanol (C24), hexacosanol (C26), octacosanol (C28) and triacontanol (C30), and phytosterol standards, including campesterol, stigmasterol, β-sitosterol, sitostanol, and 5α-cholestane, were purchased from Sigma-Aldrich (St. Louis, MO, USA). N,O-bis(trimethylsilyl)-trifluoroacetamide (BSTFA) with 1% trimethylchlorosilane (TMCS) and pyrogallol were also purchased from Sigma-Aldrich (St. Louis, MO, USA).

Enzymatic Synthesis of Lipid Esters

Comprehensive Characterization of FMBS, FMPB, and FMMB

FMSB, FMPB and FMMB (mixture of FMBS and FMPB) used in this research were supplied by the market of Qinshui Country (Shanxi Province, China) . Prior to analysis, all samples were dried, ground to powder, passed through a 60-mesh screen, and then stored in a refrigerator until analyzed.
Individual standards of fatty acid methyl esters (FAMEs, purity ≥ 99%) including methyl ester of lauric (12:0) , myristic (14:0) , palmitic (16:0) , palmitoleic (16:1) , stearic (18:0) , oleic (18:1) , linoleic (18:2) , linolenic (18:3) , arachidic (20:0) , eicosenoic (20:1) , behenic (22:0) , lignoceric (24:0) , 17 amino acids, 8 isoforms of tocopherols (Ts) and tocotrienols (T3s) , 5-α cholesterol standard (purity ≥ 95%) , campesterol, stigmasterol, β-sitosterol, sitostanol, squalene (purity ≥ 98%) , acetonitrile, n-hexane and methanol of chromatographic grade were all obtained from Sigma -Aldrich Co. (St. Louis, MO, USA) . Other reagents were of analytical pure grade and directly used without further purification.
