The Bicinchoninic acid (BCA) protein assay kit is a colorimetric detection and quantification method for determining the total protein concentration in a sample. The assay is based on the reduction of Cu2+ to Cu1+ by protein in an alkaline medium, and the subsequent chelation of the Cu1+ ion with bicinchoninic acid, resulting in a purple-colored reaction that absorbs light at 562 nm.
