Murine Lymphocyte Phenotyping by FACS

Anti-mouse antibodies for FACS were: PE-conjugated anti-mouse CD8a (clone 53-6.7; BD Pharmingen), AF700-conjugated anti-mouse CD44 (IM7; BD Pharmingen), e450-conjugated anti-mouse FoxP3 (FJK-16s, eBioscience), anti-CD3e (500A2; Biolegend), anti-CD8a (5H10; Caltag), anti-CD4 (RM4-5; BD Pharmingen), V450-conjugated CD45.1 (A20; BD Biosciences), PE-Cy7 conjugated anti-CD45.2 (104; BD Biosciences) and anti-CD25 (Clone PC61; BD Pharmingen). Functional grade anti-CD3 (17A2) and anti-CD28 (37.51) were purchased from eBioscience. For FACS cells were blocked with anti-mouse FcgRIII/IIR (BD Pharmingen) for 10 m and then stained for 45 m on ice with fluorophore-conjugated antibody. Stained cells were washed, fixed with 3% paraformaldehyde and analyzed on LSRII instrument with CellQuest (BD Biosciences) software. Data analyses were performed with FlowJo software (version 8.73; Tree Star).

Multiparametric Flow Cytometry Analysis

Flow cytometry analysis was performed by Attune Acoustic Focusing Cytometer (Life Technologies) using FlowJo software (Tree Star). For Intracellular cytokine staining, cells were stimulated with 20 ng/mL phorbol 12‐myristate 13‐acetate (Sigma) and 1 mmol/L ionomycin (Sigma) for 5 hour in the presence of a GolgiStop (BD Bioscience). The antibodies used were as follows; anti‐CD16/CD32 (clone 2.4G2; BD Bioscience), anti‐CD4 (clone H129.19; BD Bioscience), anti‐CD25 (clone PC61; BD Bioscience), anti‐CD103 (clone M290; BD Bioscience), anti‐GITR (clone DTA1; BD Bioscience), anti‐CTLA‐4 (clone UC10; BD Bioscience), anti‐Foxp3 (clone FJK‐16s; eBioscience), anti‐CD11c (clone HL3; BD Bioscience), anti‐CD80 (clone 16‐10A1; BD Bioscience), anti‐CD86 (clone GL1; BD Bioscience), anti‐CD49b (clone HMa2; BD Bioscience), anti‐LAG3 (clone C9B7W; BD Bioscience), anti‐CD11b (clone M1/70; BD Bioscience), anti‐Ly6C (clone AL‐21; BD Bioscience), anti‐CD115 (clone AFS98; eBioscience), anti‐F4/80 (clone BM8; eBioscience), anti‐CD206 (clone C068C2; BioLegend), anti‐IFNγ (clone XMG1.2; eBioscience), anti‐IL‐4 (clone BVD4‐1D11; eBioscience), anti‐IL‐10 (clone JES5‐16E3; eBioscience) and isotype‐matched control antibodies.

Identification and Quantification of ILC2 in BALF

Cells were analyzed by flow cytometry following staining with live/dead aqua dyes (Live/Dead fixable aqua dead cell stain kit, Invitrogen, 1/1000) and antibodies specific for the following markers: anti-TCRβ (clone H57-597, 5.0 μg/ml) from BioLegend; anti-CD45R/B220 (clone RA3-6B2, 1.0 μg/ml), anti-CD2 (clone RM2-5, 0.8 μg/ml), anti-CD25 (clone PC61, 1.0 μg/ml), anti-CD90.2 (clone 53-2.1, 2.5 μg/ml), and anti-CD11b (clone M1/70, 0.3 μg/ml) antibodies from BD Pharmingen; anti-CD45.2 (clone 104, 0.5 μg/ml) from eBioscience. ILC2 were gated as CD45+, CD2 TCRb B220 CD11b, CD90+ and CD25+ cells in a similar manner to that described by Roediger et al.10 (link). Bronchoalveolar lavage fluid (BALF) cells were additionally labeled using anti-Siglec F (clone E50-2440, 2.0 μg/ml), from BD Pharmingen; anti-Gr-1 (clone RB6-8C5, 0.4 μg/ml), anti-CD8 (clone 53-6.7, 5.0 μg/ml), anti-CD4 (clone GK1.5, 1.0 μg/ml), anti-CD3 (clone 2C11, 1.0 μg/ml) from eBioscience; and anti-CD11c (clone N418, 2.5 μg/ml) from BioLegend.
