Screening of 56 Antischistosomal Compounds

A total of 56 compounds (Figure S4, Table S4) was selected by both the manual and automated methods for screening
of S. mansoni adults and somules. The following
compounds were purchased from Cayman Chemical: U-73122, BIX01294 hydrochloride
hydrate, tyrphostin AG-1478, sivelestat sodium tetrahydrate, AGK2,
trilostane, itraconazole, dabigatran etexilate, SB202190 hydrochloride,
niflumic acid, amsacrine hydrochloride, R(+)-IAA-94,
piperlongumine, tiamulin fumarate, PNU-282987, tetrabenazine, nemadipine-A,
mycophenolic acid, etravirine, (S)-duloxetine hydrochloride,
moxidectin, and rutecarpine. The following compounds were purchased
from Sigma-Aldrich: clindamycin 2-phosphate, tyrphostin I-OMe-AG-538,
and escabet sodium. The following compounds were purchased from Selleck
Chemicals: revaprazan hydrochloride and Org 27569. The following compounds
were purchased from Enamine: Z385159220, Z18885599, Z276431168, Z2241105867,
Z827016000, Z48867676, Z53005631, Z90192490, Z56174662, Z105384660,
Z44528364, Z827015296, Z304863612, and Z56958732. Finally, the following
compounds were purchased from Toronto Research Chemical: eletriptan
and ondansetron hydrochloride dihydrate. Powders were stored according
to vendor specifications. Compounds were then dissolved at 10 mM in
fresh DMSO and shipped to UCSD on dry ice for storage at −80
°C until use.

Evaluating HIV-1 Infection in CD4 T Cells

Isolation of resting CD4 T cells from uninfected subjects’ PBMC (Lifesource, Rosemont, IL), culture, preparation of virus stocks, virus infection, qPCR for HIV-1 DNA were performed as described elsewhere (Taylor et al., 2015 (link)). Single-round CXCR4-tropic envelope-pseudo-typed GFP-expressing HIV-1 (Zhou et al., 2005 (link)) and HIV-1 NL4-3, a CXCR4-tropic virus, were used specifically to obviate any possible effect of mTORi on cell CCR5 expression (Heredia et al., 2003 (link)). Flow cytometric analysis of single-round infections allowed a quantitative analysis of viral replication over a 4-log dynamic range (Zhang et al., 2004 (link)). Where indicated, cells were treated with cytokines or chemokines (Peprotech) at 100 ng/ml, or pretreated with VU0359595 (PLDi) (10, 2, or 0.4 μM, Cayman Chemical), rapamycin (1, 0.2, or 0.04 μM, Cayman Chemical), AZD2014 (mTORi #1) (5, 1, 0.2 μM, Selleckchem), AZD8055 (mTORi #2) (5, 1, 0.2 μM, Selleckchem), TAK-228 (mTORi #3) (5, 1, 0.2 μM, Cayman Chemical), mycophenolic acid (10 μM, Cayman Chemical), or treated with indicated nucleoside (Sigma) at 50 μM at the time of stimulation.

In Vitro Iohexol Elimination Assay

Sterile sodium chloride (0.9% NaCl) and phosphate-buffered saline (PBS) were purchased from Baxter Healthcare Corporation (Deerfield, IL). Iohexol powder, paclitaxel, and gemcitabine were purchased from Toronto Research Chemicals Inc (Toronto, ON, Canada). Iohexol dosing solution was manufactured by GE Healthcare, (Shanghai, China) and obtained from the Hillman Cancer Research Pharmacy. All in vitro drug elimination incubations with iohexol were performed at 300 μg/mL which is at the upper range of the reported plasma concentrations [23 (link)]. [D5]-iohexol (IS) was purchased from ALSACHIM (Graffenstaden, France). Carboplatin for dosing was manufactured by Ben Venue Labs Inc (Bedford, OH) and obtained from the Hillman Cancer Research Pharmacy. NADPH tetrasodium salt was purchased from Enzo Life Sciences (Farmingdale, NY). Carboplatin powder, MgCl2, alamethicin, UDPGA sodium salt, phenacetin, dextromethorphan and midazolam methanolic solution was obtained from Sigma (St Louis, MO). [D3]-dextrorphan and [D4]-1-hydroxy midazolam were purchased from Sigma (St. Louis MO). 1-hydroxy midazolam, 4-hydroxy diclofenac, 4-hydroxy mephenytoin, dextrorphan tartrate, s-mephenytoin, SN-38, trifluoperazine, mycophenolic acid and naloxone were purchased from Cayman chemicals (Ann Arbor, MI).
