Quantitative Analysis of c-di-AMP Levels in S. pneumoniae

S. pneumoniae strains used in this study were derived from D39 Δcps (58 (link)) unless otherwise stated. S. pneumoniae cells were grown in Todd Hewitt broth containing 0.5% yeast extract at 37 °C with 5% CO2. Strains, plasmids, and oligonucleotides used in this study are given in SI Appendix, Table S1. The c-di-AMP levels were quantified using a “Cyclic-di-AMP ELISA Kit” from Cayman Chemical (item No. 501960) in accordance with the manufacturer’s instructions. Quantifying Cyclic di-AMP in Infected Cells

HeLa cells were infected with OncoVACV or STINGPOX (MOI = 0.1) and 48 hours post-infection cells and supernatants were harvested. Cells were lysed in mammalian protein extraction reagent (M-PER; Thermo Scientific) and boiled at 95°C for 5 minutes. Cyclic di-AMP levels were measured in the cell lysate and supernatant via the cyclic di-AMP ELISA kit (Cayman Chemicals), as per manufacturer’s protocol.
